| Objective:To observe the effect of Yanlurukang Capsule on the breast tissue,estrogen and progesterone,expressions of estrogen receptor and progesterone receptor in rats with hyperplasia of mammary glands(HMG),discuss the therapeutic mechanism of HMG.In addition,also,provide experimental basis for the clinical application of the drug.Methods:Combined Estradiol benzoate with progesterone by intramuscular injection to copy breast Hyperplasia rats model.50 nonfertile female healthy adult Wistar rats were randomly divided into normal group,disease model group,the Yanlurukang Capsule group and Tamoxifen group,including 15 rats of the model group and 15 rats of the normal group,remaining 10 rats in each group.After adaptive feed for a week,exceptforthenormalgroup,therats inother5 groupsweretreatedwith benzoate estradiol(0.5mg/kg)and progesterone(4mg/kg)subcutaneous injection to duplicate the model of hyperplasia of mammary glands.After modeling success,the rats in normal group and disease model group were received normal saline,the rats in other groups were respectively administrated by corresponding medicines,all for 30 days.To observe rats general condition,changes in the nipple diameter and height.Determination the concentrations of estradiol(E2),progesterone(P)in rats serum.Observe the changes of each group breast tissue in rats.Detect the expressions of ER and PR in breast tissues by the method of immunohistochemical staining(IHC).Results:1.Changes in nipple diameter and height:compared with the normal group,the height and diameter of nipples in disease model group were increased(P<0.05);Compared with model group,the height and diameter of nipples in Yanlurukang Capsule group and Tamoxifen group were reduced markedly(P<0.01).1.The rat mammary glands pathology:Compared with the normal group,the model group rats mammary glands tissue ductal epithelial layer and acinar number increasedsignificantly(P<0.01),part of the structure of mammary gland disorder,present hyperplasia change.Compared with the model group,the model group rats mammary glands tissue ductal epithelial layer and acinar number were lowered remarkably(P<0.01),duct hyperplasia of mammary glands and breast ducts cavity secretions were also improved.3.The level of E2 and P:Compared with the normal group,the level of E2 and were increased remarkably(P<0.01),but the concentration of P were were decreased markedly(P<0.01).Compared with the model group,the level of E2 was decreased evidently in Yanlurukang Capsule group and Tamoxifen group(P<0.01),but increase the level of P obviously(P<0.01).4.The expression of ER and PR:Compared with the normal group,the disease model group ER and PR level were increased remarkably in breast tissue(P<0.01);Compared with the model group,the expression of ER and PR reduced significantly in Yanlurukang Capsule group and Tamoxifen group(P<0.01).Conclusion:Yanlurukang Capsule can reduce the diameter and height of the rats nipple,inhibit the excessive secretion of E2,increase the level of P of rats serum and reduce the expression of ER and PR significantly.Furthermore,it also can improve the pathological morphology of rat with hyperplasia of mammary glands.Yanlurukang Capsule have a good protective and the therapeutic action on hyperplasia of mammary glands,its mechanism may be related to regulating endocrine hormone disorder and reducing the expressionofestradiolreceptor(ER)andprogesteronereceptor(PR). |
