Objective:The target of this study was to investigate the effect of PES on the proliferation,migration and invasion of gastric cancer SGC-7901 cell line and its mechanism.Methods: The logarithmic phase cells were divided into four groups: 37?DMSO group,37?PES group,43?DMSO group and 43?PES group.Each group was cultured for 1 hours under the condition of the experiment,and then continued to culture under the condition of 37?,5%CO2 cell culture medium:1.The expression of HSP70 in each group was detected by Western blotting after cultured 6 hours later;2.CCK-8 assay to evaluate the proliferation of cells in each group on the point of 24 h,48h and 72 h.3.In addition,Transwell chamber assay was used to examine the migration and invasion of SGC-7901 cells.Results: 1.West-blot results showed that the gastric cancer SGC-7901 cells can express HSP70 protein,besides,the HSP70 protein was over-expressed by gastric cancer SGC-7901 cells under the condition of hyperthermia(P<0.05)(group 43?DMSO VS group 37?DMSO :2.28±0.06 VS 1.52±0.01).Meanwhile,PES can decline the expression of HSP70 protein in SGC-7901 cells at the temperature both 37 and 43? ?(P<0.05)(group 37?PES VS group 37?DMS : 0.76±0.02 VS 1.52±0.01 and group 43?PES VS group 37?DMSO :1.25±0.05 VS 1.52±0.01).2.CCK-8 experiment showed that the group 37?PES,group 43?DMSO and group 43?PES all can inhibit the proliferation of the SGC-7901 cells compared to the group 37?DMSO after treated(P<0.05),and the inhibition effect of the group 43?PES was the most obvious(P<0.01).3.Transwell migration assay showed the migration numbers of the cells after treated 24h:the group 43?DMSO(40.40±2.70),the group 37?PES(25.40±2.97)and the group 43?PES(17.20±1.48),compared to the group 37?DMSO(58.80±2.86),the former three groups can all inhibit the migration of the SGC-7901 cells after treated(P<0.05),and the group 43?PES showed the most obvious inhibition effect(P<0.01).Transwell invasion assay showed the invasion numbers of the cells after treated 24h:the group 43?DMSO(28.00±2.45),the group 37?PES(17.60±1.95)and the group 43?PES(11.60±1.95),compared to the group 37?DMSO(38.00±4.36),the former three groups can all inhibit the invasion of the SGC-7901 cells after treated(P<0.05),and the group 43?PES showed the most obvious inhibition effect(P<0.01).Conclusion:PES can inhibit the proliferation,migration and invasion of gastric cancer SGC-7901 cells under the condition of hyperthermia,and its mechanism may be related to the inhibition of HSP70 protein expression in gastric cancer SGC-7901 cells by PES.
