Preventive Effects Of Jianpi Huoxue Decoction On Nsaids-induced Injury To The Mucosa Of The Small Intestine In Rats

Objective: To establish the model of NSAIDs enteropathy induced by diclofenac and investigate the effect of Jianpi Huoxue Decoction on small intestinal injury,the intestinal pathological results,permeability in intestinal,the influence of the TNF-α.To provide experimental evidence for clinical prevention and treatment of NSAIDs related intestinal injury.Methods: Forty-eight SD rats adaptive feeding for 7 days,were randomly divided into six groups for eight rats in each group including negative control group,diclofenac model group,glutamine treatment group,Jianpi Huoxue Decoction treatment of low,medium and high dose groups.Both model group and treatment groups were given diclofenac 9.89 mg/kg/d.Four treatment groups were given glutamine,Jianpi Huoxue Decoction of high(15.12 g/kg/d),medium(7.56 g/kg/d),low(3.78 g/kg/d)dose half an hour in advance.1 ml/100g/d distilled water was given to rats in the blank control group for 7 days.12 hours after the last administration,anesthesia by intraperitoneal injection of 4% chloral hydrate.Take inferior vena venous blood to detect D-lactate,TNF-α Levels.Observe the entire section of the small intestine.Take small intestine tissue from the ileocecal 40 cm proximal side to take intestinal 1cm for pathological sections and to observe the changes in form and pathology.Results:(1)Establishment of NSAIDs related intestinal injury model.Different doses of diclofenac were found that with the increase in dose and time,the intestinal mucosal damage significantly increased.To select diclofenac 9.89 mg/kg/d for 7 days as an experimental dose can appear intestinal mucosal injury performance,and the general condition of rats can still be successful modeling.(2)The protective effect of Jianpi Huoxue Decoction on NSAIDs enteropathy(1)Compared with the negative control group,the rats in the model group were generally poor,fur fluffy,apathetic,unresponsive,slow weight gain,significantly reduced food intake,dilute stool.The rats in the treatment group were still in general,and the rats in the high dose group were the best.(2)Compared with the blank group,the intestinal mucosa of the model group showed obvious congestion and edema,erosion,ulcer.One of them can be seen severe intestinal swelling,local intestinal necrosis.In the treatment group,the small intestine of glutamine group showed local congestion and edema,and the degree of intestinal ulcer was less than that of model group.Jianpi Huoxue Decoction groups of rats were seen mucosal congestion and edema,low dose occasional small ulcers.The degree of intestinal injury was significantly lower than that of model group.Compared with the normal group,the pathological section of the model group showed that the small intestine tissue structure was obviously damaged,some mucosal necrosis was lost and the lesion reached the submucosal layer.Compared with the model group,the damage to treatment group was significantly reduced.(3)Compared with normal group,the D-lactic content increased significantly in model group(P<0.01);Compared with model group,The levels of D-lactic in glutamine group and Jianpi Huoxue Decoction group were significantly decreased(P< 0.05).The most significant reduction in the high dose group was statistically significant(P<0.01).(4)Compared with normal group,the TNF-α level in the model group was significantly higher(P<0.01);Compared with the model group,the levels of TNF-α in glutamine group and Jianpi Huoxue Decoction group were significantly lower(P < 0.05).The most significant reduction in the high dose group was statistically significant(P<0.01).Conclusion: Jianpi Huoxue Decoction can improve the general condition of rats,prevent and treat NSAIDs-induced Injury to the Mucosa of the Small Intestine.To reduce the occurrence of erosion,ulcer and pathological damage and improve the intestinal mucosal permeability,reduce inflammation.
