ObjectiveTo explore the effect on the proliferation and apoptosis of human ovarian cancer SKOV3 cells induced by celastrol.and further study the role of reactive oxygen species(ROS)in the apoptosis induced by celastrol in SKOV3 cells.It provided a theoretical basis for the treatment and prognosis of celastrol for ovarian cancer.MethodsThe effect of celastrol on inhibiting SKOV3 proliferation was determined by MTT staining method;The intracellular ROS levels were detected by flow cytometry arespectively;Hoechst 33258 staining and flow cytometry were used to detect the effects of apoptosis,and the effect of apoptotic proteins was determined by Western blot method;NAC was used to observe the role of ROS in the antitumor effect of celastrol.ResultsCelastrol had obvious proliferation inhibitory effect on ovarian cancer SKOV3 cells,and showed a time/dose dependence relationship;Celastrol induced significant apoptosis in ovarian cancer SKOV3 cells;Celastrol can significantly increase intracellular ROS levels in SKOV3 cells,and up-regulated the expression of cleaved caspase-3 and cleaved caspase-9,and down-regulated the expression of anti-apoptotic protein bcl-2;NAC antagonized the antitumor effect induced by celastrol.ConclusionsCelastrol induces apoptosis in human ovarian cancer SKOV3 cells by increasing intracellular ROS levels.
