Objective:To study the effects of eletroacupuncture at Changqiang(GV1)acupoint on CREB expression in the multiple brain region of the FMR1 knockout mice.Methods:A total of twenty-four 48-days FMR1 knockout mices were randomly divided into blank group,a eletroacupuncture at a non-acupoint group,a eletroacupuncture at Changqiang(GV1)and Each group of eight The Changqiang(GV1)of the mice is The hollow between the tail and the anus;eletroacupuncture at a non-acupoint group is located at 1 cm from the lowest point of the right arch of the mice;The blank group was reared under the same conditions without any intervention.The eletroacupunture apparatus was connected with continuous wave,2 mA,2 Hz,20 min,once a day,seven times a week for two weeks.After 14 days of continuous intervention,the mice were drawn on the 15th day.All the groups were tested before and after intervention to detect the feces number and activity.Cortical area,cerebellar area and hippocampal tissue were collected after intervention.The expression of CREB,ERK and p-CREB protein was detected by immunohistochemistry and Western blot.Results by comparing and analyzing the data with SPSS 22.0 software,the statistical method to choose the single factor analysis of variance method(one-way ANOVA),data are for double check,P<0.05,explain the differences statistically significant,finally using Graphpad Prism statistical graphics.Results:(1)The test results of the independent behavior showed that a eletroacupuncture at Changqiang(GV1)group was compared with that of the blank group and a eletroacupuncture at a non-acupoint group,The activity level increased,the activity level increased,and there was a significant difference(P<0.05).(2)The results of immunohistochemistry of brain regions showed that in the cortical area,the average optical density of CREB positive cells in the eletroacupuncture at Changqiang(GV1)group was higher than that in the blank group,with significant difference(P<0.05).The average optical density of p-CREB positive cells in the three groups was relatively average,with no significant difference(p>0.05).At the same time,the average optical density value of ERK positive cells in the eletroacupuncture at a non-acupoint group was lower than that in the blank group and the eletroacupuncture at Changqiang(GVI)group,with significant difference(P<0.05).In the cerebellum area,the average optical density values of CREB and p-CREB positive cells in the three groups of cerebellum were average,and there was no significant difference(p>0.05).The average optical density of ERK positive cells in the eletroacupuncture at Changqiang(GV1)group was higher than that in the eletroacupuncture at a non-acupoint group,and there was a significant difference(P<0.05).The average optical density of CREB positive cells in the blank group of the hippocampus was higher than that in the eletroacupuncture at Changqiang(GV1)group and the eletroacupuncture at a non-acupoint group,and there was a significant difference(P<0.05).The average optical density values of ERK and p-CREB positive cells in the three groups in the hippocampus were average,no significant difference(p>0.05).(3)The results of Western blot test showed in the cortical region,compared with the blank group,there was a significant difference between the eletroacupuncture at a non-acupoint group and the eletroacupuncture at Changqiang(GV1)group about CREB protein expression(P<0.05).At the same time,the ERK and p-CREB protein expression of the eletroacupuncture at Changqiang(GV1)group was higher than that in the blank group and the eletroacupuncture at a non-acupoint group,with significant difference(p<0.05).The CREB and ERK protein expression of the eletroacupuncture at Changqiang(GV1)group in the cerebellar area was higher than that in the blank group and the eletroacupuncture at a non-acupoint group,with significant difference(P<0.05).There was no significant difference in p-CREB protein expression between the three groups(p>0.05).The CREB protein expression in the eletroacupuncture at a non-acupoint group in the hippocampus was lower than that in the blank group and the eletroacupuncture at Changqiang(GV1)group,with significant difference(P<0.05).The ERK protein and p-CREB protein expression in the blank group were higher than that in the eletroacupuncture at Changqiang(GV1)group and the eletroacupuncture at a non-acupoint group,and there was a significant difference(p<0.05).Conclusion:(1)Effects of eletroacupuncture at Changqiang(GV1)acupoint can increase the expression of CREB and ERK in the cortical region and cerebellum region associated with exercise,and the results are consistent with the behavioral results.(2)Eletroacupuncture at Changqiang(GV1)acupoint in multiple brain regions,can strengthen the cortical region and cerebellar area the linkage effect,and the expression of CREB related proteins in the hippocampus was weakened.
