Objective:To observe the effect of acupuncture at "Changqiang"(GV1)point on the expression of NF-κB and Bcl-xL proteinin the hippocampus of autism model rats.Methods:(1)The healthy adult Wistar rats of SPF grade were caged with male and female 1:1,and the rat tray was checked on the morning of the next day.If translucent vaginal plug was found in the tray,it was recorded as 0.5 day,confirm pregnant females kept separately;(2)The pregnant mice were randomly divided into two groups according to the random number table.One group of pregnant rats were injected intraperitoneal injection of sodium valproic acid for 12.5 days,On behalf of autism model rats,the birth of mice in model group;another group of pregnant rats in E12.5 intraperitoneal injection of normal saline,and birth to the blank group;(3)The offspring of offspring were subjected to developmental condition,forced swimming,hot plate causing pain,and open field behavior tests to screen the offspring that fit autism model;(4)From the twenty-four model group rats were randomly selected from the model group,and were randomly divided into model group,non-acupoint group and "Changqiang" point group,each group of eight,randomly selected from the blank group were blank group,From randomly selected eight rats in blank group rats into blank group."Changqiang" group take "changqiang" point,that is,between the anus and the root depression position,started to intervene in the treatment in rats 24 days of age,straight piercing,needle depth of about 0.3-0.4 inch,line mentioning method,continuous needle 1 min,the frequency of 160-200 times/min.The non-acupoint group was fixed on the right side with non-acupoints and the remaining operation was the same as that of the "Changqiang" group.The model group and the blank group were housed in the same environment and were simulated and grasped without acupuncture;(5)All groups were subjected to open-field behavioral testing before intervention(23 days old)and after intervention(34 days old),and tissue in hippocampus of rats was taken on the 34th day.The expression of NF-κB and Bcl-xL in the hippocampus was detected by Western blotting;(6)All the data are processed spss20.0 software analysis.Results:(1)Body weight index test:Compared with the blank group,the body weight of the model group increased slowly,PN7 had no significant difference(P>0.05),PN14、PN21、PN35 had significant difference(P<0.05).(2)Eyes open experiment:compared with the blank group,the eyes of VPA autism model group had a longer open-eye time.There were significant differences in PN14,PN15 and PN16(P<0.05)and no significant difference in PN13 and PN17(P>0.05).(3)Forced swimming test:compared with the blank group,the VPA autistic model rats swimming ability was significantly lower in the PN9,PN11,PN13,PN15 and other age groups were significantly different(P<0.05).(4)Hot plate pain test:compared with the blank group,the VPA autistic model offspring pain obvious slowness,showed a significant increase in pain threshold,escape nociceptive stimuli more slowly.There was significant difference(P<0.05)in the pain-inducing time of PN9,PN11,PN13 and PN15 between the two groups with statistical significance.(5)Open field behavior:1)before intervention:compared with the blank group,the total distance of the model group,the "Changqiang" group,the non-acupoint group was shorter,the average speed was slower,the activities in the peripheral area were longer,the repeated stereotyped movements were more,There was a significant difference between the group and the other three groups in the total distance,average speed,activities time in the peripheral area and repetitive stereotyped movements(P<0.05);2)after intervention,the total length ofthe"Changqiang" group increased significantly compared with the previous group,the average speed increased,the activity time in the peripheral region shortened and the repeated stereotyped motion decreased.Compared with the blank group,the total distance,average speed,(P>0.05).Compared with the model group and the non-acupoint group,the total distance,average speed,peripheral time and repeated stereotypes were significantly different(P<0.05).(6)Western Blot method:compared with the blank group,the protein bands of NF-kappa B and Bcl-xL in the hippocampus of VPA autism model group and non-acupoint group were obviously increased,and the quantitative analysis showed that the protein expression level of model group decreased significantly(P<0.05).The relative volume of NF-kappa B and Bcl-xL protein bands in hippocampus of the "Changqiang" group was similar to that of blank group,and the quantitative analysis showed that there was no significant difference(P>0.05).Conclusion:1.Acupuncture at "Changqiang" acupoint can improve abnormal behavior pattern of autism rats.2.The expression of NF-kappa B and Bcl-xL protein in hippocampus of the autistic model rats was up-regulated compared with the blank group.3.Acupuncture "Changqiang" acupoint can down-regulate the expression of the NF-kappa B and Bcl-xL protein in hippocampus of VPA autism model rats.
