| Objective:To study the traditional Chinese medicine Xin Wen Tongyang fistular onion stalk extract on protective effect and mechanism of myocardium in rats with heart failure after myocardial infarction.Methods:50 male SPF SD rats by ligation of the left coronary artery were randomly divided into blank model group,perindopril group,onion extract group,another 10 in line,without ligation,feeding one week,perindopril group,fistular onion stalk extract group which were treated with 2mg/(kg·d)the corresponding drugs by gavage,saline sham operation group and blank model group to be given orally administered continuously for 12 weeks,during the growth of rats,determination of rat heart/body weight index,the left ventricle of rat HE and Masson staining and ELISA method for the determination of Qing Dynasty BNP、IL-6、TNF-α、AngII、TGF-β1 in rat blood analysis using real-time fluorescence quantitative(RT-qPCR)and Western blot technique(Western blot)expression in myocardial tissue of rats was detected in Col I-mRNA,Col-mRNA、protein Col I and III.Results:the sham operation group,shiny coat,a good mental state;the blank model group rats were dispirited,haggard fur without light;the model group gradually recovered after drug intervention,rats food intake gradually increased,fur was smooth.In the sham operation group,the myocardial cell gap was clear,and the myocardial fibers were arranged orderly.The gap between the myocardial cells in the blank model group was different,most of the gaps were narrowed,the myocardial fibers arranged in disorder,and a large number of inflammatory cells infiltrated.The pathological changes of the drug group were relatively mild,and the intercellular space was relatively uniform compared with the model group,and the arrangement of myocardial fibers was relatively neat and the inflammatory cells were less.Compared with the sham operation group,model group rats heart weight index,serum BNP、IL-6、TNF-α、AngII、TGF-β1 was significantly increased,the difference was statistically significant(P<0.01);compared with the blank model group,perindopril group and onion extract rats heart weight index,serum BNP、IL-6、TNF-α、AngII、TGF-β1 was decreased,the difference was statistically significant(P<0.01)。Comparison between drug group,changes of heart weight index,serum BNP,AngII,TGF-β1 in rats,no difference was statistically significant(P>0.05);compared with the sham group,the expression of myocardial Col in rats in the blank model group of-mRNA,Col I,Col III-mRNA I-protein content significantly increased,the difference was statistically significant(P<0.01);compared with the model group,decreased expression levels of perindopril group and onion extract rats in myocardium of rats with ColI-mRNA,Col-mRNA,ColI-proteinIII,the difference was statistically significant(P<0.01);drug groups.Between the equivalent content of expression of Col I and ColI-mRNA/protein,ColIII-mRNA in the myocardium,there was no statistically significant difference(P>0.05).Conclusion:with the growth of onion extract in rats with heart failure after myocardial infarction improved,has protective effect on heart failure rats by deposition decreased collagen fibers in myocardium,changing the ratio of ColI/ColIII,reduce myocardial dimension in the degree of heart failure rats,its mechanism may be the inhibition of AngⅡ-AT1R-TGF-β1 signaling pathway and reverse ventricular remodeling may be related to the role of yang. |
