| Background and ObjectiveScarring of the skin is a malignancy of the skin resulting from the malignant transformation of the scar.The skin scar cancer can easily be mistaken for chronic ulcers and ignored by patients.When patients visit,the tumors have distant metastases and they are not sensitive to radiotherapy and chemotherapy.Effective results can be obtained through surgical resection for early skin scar cancers.However,for advanced or late scar cancer,the effect of surgical treatment is poor,as well as the prognosis.In recent years,many studies have demonstrated that Notch signaling pathway is involved in proliferation,differentiation and apoptosis of many kind of tumor cells.Many studies have found that Notch1 proteins play different roles in different tumors.However,the role of Notch1 in cutaneous scarring is unclear.It needs further in-depth discussion.In order to clarify the role of the Notch1 signaling pathway in normal skin,studies of hypertrophic scar and scar cancer,searches for the molecular biological labels,the early diagnosis and advanced gene therapy,have been examined the expression of Notch1 protein in skin cancer and normal adjacent tissues.We also studied the relationship between the expressions of the Notch1 protein and the pathological features of skin scars.It is to provide a theoretical basis for clinical diagnosis and treatment of hypertrophic scars and scarring skin cancer.Materials and Methods1.Immunohistochemistry(SP)method was used to detect the expression of Notch1 protein in the tissues of 47 normal skin tissues,56 hypertrophic scars and 83 cases of skin scar cancer(71 cases of phosphorous cell carcinoma and 12 cases of basal cell carcinoma)and adjacent normal tissues.2.The data obtained were analyzed by SPSSl7.0 and chi-square test.The level of α<0.05 was the test level,and P<0.05 was considered to be statistically significant.Results1.The positive expression rate of Notch1 protein in normal skin was 72.34%.The positive expression rate of Notch1 protein in hypertrophic scar was 92.86%.The positive expression rate of Notch1 protein in skin scar cancer was 7.23%.The positive rates of the three were significantly different(P<0.05).The comparison between the three groups showed that the expression of Notch1 protein in hypertrophic scars was significantly higher than that of normal skin and skin scars.In normal skin,it was higher than skin scar cancer,and the difference was statistically significant(P<0.05).2.There is a certain difference between the positive expression rate of Notch1 protein and the pathological type of tumor.The positive rate of the expression of Notch1 protein in the pathological type of squamous cell carcinoma is 7.04%,in the pathological type of squamous cell carcinoma scar cancer,the positive expression rate was 8.33%,but the difference was not statistically significant(P>0.05);and the positive expression rate was 7.81% in stage I-grade skin scar cancer,II-Ⅳ skin cancer positive The expression rate was 5.26%,the difference was not statistically significant(P>0.05).There may be no correlation between the positive rate of Notch1 protein and lymph node metastasis,and the difference is not statistically significant(P=0.944).Conclusion1.Notch1 protein may play an inhibitory role in the formation of skin scar cancer.2.Notch1 protein promotes the formation of hypertrophic scars by stimulating hyperkeratosis on the surface of the scar.3.The expression of Notch1 protein in cutaneous scar tissue may not be related to the grade of tumor,whether the tumor is associated with lymph node metastasis and the pathological type of tumor. |
