Research Of Analytical Method And Prevention Technique Against Aflatoxins And Aspergillus Flavus In Semen Nelumbinis And Radix Notoginseng

ObjectiveThis study evaluated the growth inhibition and antiaflatoxigenic effect of essential oil and related preparation and “medicine couple method”.Then,these two technologies were used as the storage protection tools for usual medicine Semen Nelumbinis and rare medicine Radix Notoginseng to prevent from the infection of Aspergillus flavus and aflatoxins productions.MethodsThis study developed a HPLC-FLD coupled with IAC clean-up method for aflatoxins detection in Semen Nelumbinis and Radix Notoginseng.A fumigation method was used to evaluate the anti-Aspergillus flavus effect of 12 essential oil and prepared the four most efficient oil to gel and film agent and test again.Finally,the most efficient preparations were used on the protection of Semen Nelumbinis to assess the actual application effect.A trans-inoculation test was used to evaluate the anti-Aspergillus flavus effect of 9 herbs,then coupled the effective herbs with Radix Notoginseng and incubated in a high temperature and high humidity condition.Finally,the most efficient protector medicine and optimal dosage were given.ResultsAfter screening,the cinnamon essential oil,Litsea cubeba essential oil,Palmarosa essential oil and Thyme essential oil had an efficient antifungal ability to Aspergillus flavus.Then,these 4 essential oils were prepared to gel and film agents and the antifungal effects were decline,and there were also a good effect in a high concentration.Finally,the cinnamon essential oil gel agent and Palmarosa essential oil film agent were as two efficient antifungal agent for the protection of Semen Nelumbinis.After screening,the Cortex Cinnamomi,Fructus Anisi Stellati,Cortex Moutan and Flos Syzygii Aromatici had an efficient antifungal ability to Aspergillus flavus.After coupling these 4 herbs with Radix Notoginseng and incubation,the Cortex Moutan was the optimal protector agent and the dosage ratio was 2:1.ConclusionsThis study successfully screened two essential oil preparations and a protector medicine for effectively inhibiting the growth of Aspergillus flavus by antifungal test and trans-inoculation test.And it can support a satisfactory reference to the common or large-scale herbal warehouse.
