| In the process of collection,processing,transportation and storage,traditional Chinese medicine may be infected by a variety of fungi,especially toxic fungi,due to the influence of their own factors and environmental conditions,this may produce mycotoxins with serious side effect,which will affect the quality and safety of traditional Chinese medicine and its products.Mycotoxins are not showing acute toxicity to human body,but they have strong accumulation in human body.It will damage the function of liver and kidney,pose serious threat to humanhealth,even the safety of life.There are many kinds of traditional Chinese medicine and their mildew condition are also different,the contamination and residual level of mycotoxins and their influence on the quality of medicinal materials are also quite different.Methanol-water,acetonitrile-water were used as extraction solvents for the detection of mycotoxins with good liposolubility,while the conventional medication form of traditional Chinese medicine materials is water decoction.Therefore,establish a method for the detection of multi-mycotoxins in easily moldy traditional Chinese medicines,study on the residue and accumulation of mycotoxin in different traditional Chinese medicine and its transfer in the decoction,and explain the effect of mycotoxin contamination on the quality of medicinal materials are of great significance to ensure the quality of medicinal materials and the safety of users.This paper research content mainly has the following several aspects:(1)Establish a reliable UFLC-MS/MS method for the detection of 18 mycotoxins.After the preparation of the extract from the five kinds of cultured traditional Chinese medicines such as Lilii Bulbus,Hordei Fructus Germinatus,Nelumbinis Semen,Polygalae Radix and Bombyx Batryticatus,the mycotoxins will be detected by the established method,to determine the toxin production of the five kinds of traditional Chinese medicine,and the results showed that the method established is accurate and reliable,the toxin production are significantly different and the amount of aflatoxins are the most.(2)To investigate the transfer of aflatoxins in the five kinds of traditional Chinese medicines.They were cultured in mold incubator(humidity:90%;temperature:28℃)for 10 days,after extracting the aflatoxins of the original medicinal materials and there decoctions by immunoaffinity column purification,a sensitive post-column derivatization-HPLC-FLD method for the detection of aflatoxins in the original medicinal materials and there decoctions was established,and the transfer of aflatoxins was determined.The results showed that the method established is accurate and reliable and the transfer rates of the five medicinal materials were quite different.(3)To investigate the effect of aflatoxins on the quality of medicinal materials.Firstly,LC-MS/MS method was established to detect four kinds of aflatoxins in five kinds of traditional Chinese medicines,and the HPLC-PDA method was used to determine its chemical composition.The five kinds of traditional Chinese medicines were cultured in mold incubator(humidity:90%;temperature:28℃)for 20 days,and take out some medicinal materials every 4 days to detect the content of aflatoxins and chemical constituents,Investigate the fungal infection of traditional Chinese medicines and the influence of accumulation of aflatoxins on its quality.The results showed that With the prolongation of culture time,the content of aflatoxin in traditional Chineses medicines gradually increased,while the content of its chemical constituents decreased,which greatly influenced the composition of most traditional Chineses medicines. |
