| Objective:To date,high-fat diet is the important inducement of the obesity..In this study,we observe the change of high-fat diet induced in mice,and then characterized the effect and mechanism of Ping tang Recipe on lipid deposition and adipose tissue function in mice induced by high-fat diet.Methods:After a week of adaptive feeding,the mice were randomly divided into the control group,the HFD group and the drug-treated group.The control group normal was given normal diet,while HFD group and drug-treated group was given diet with 60%fat feed for two weeks,before Ping tang Receipe intervention.Control group and HFD group received saline and treated group received Ping tang receipe via gastric daily gavage.After another 10 weeks,insulin resistance and glucose tolerance were measured by IPITT and IPGTT assay.Energy intake and expenditure were analysed in Comprehensive Lab Animal Monitoring System(clams)by monitoring the 24-hour O2 consumption,CO2 release and respiratory quotient(RQ)of individual mice in each group.Visceral adipose tissue(VAT),subcutaneous adipose tissue(SAT)were collected and weighted;Serum were collected and further analyzed for TG,TC,NEFA and LDL-C.HE staining was applied to compare the morphology of adipocyte in each group.Western Blot was used to detect the expression of fatty acid metabolism related enzymes and transcription factors(FAS ACC1,CPT1,PPARγ)in the visceral fat and subcutaneous adipose tissue.Immunohistochemistry at F4/80 was applied to identify the macrophages infiltration in adipose tissue,Real-time PCR was applied to quantify inflammation status and macrophage polarization in adipose tissue,including TNFa and chemokines MCP-1,Mrc and Retnla.Results:Compared to the HFD group,Ping tang Recipe treatment reduced the body weight(P<0.001)significantly.Glucose tolerance and insulin sensitivity were also ameliorated by Ping tang Recipe according to IPGTT and IPITT test.Furthermore,it was shown that Ring tang Recipe treatment could moderately increase O2 consumption and CO2 production as well as carbohydrate oxidation rate and energy consumption.RQ values of control group and HFD group was 1 and 0.7,respectively.Ping tang Recipe treatment could moderately increase the RQ to 0.8,which indicating a different reliance on carbohydrate against lipids.As compare with control group,the adipose weight of drug-treated group with a tendency to significantly reduction,and the subcutaneous fat weight,was statistically significant(P<0.05)with the plasma TG,TC,LDL-C,NEFA levels decreased(P<0.05),HE results showed the visceral fat and subcutaneous fat cell of drug-treated group was less of number,and smaller volume;Western blot results indicated that the FASN in visceral and subcutaneous adipose tissue of drug-treated group and ACC1,PPAR gamma protein expression with a trend of increase(P<0.05),and the macrophages of visceral and subcutaneous adipose tissue for control group infiltrate significantly(P<0.05),the TNF alpha,MCP-1 and CD11c mRNA expression of the drug-treated group were relatively decreased,while the Mrc,Retnla mRNA levels expression of the drug group were increased(P<0.05).Conclusion:Ping tang Recipe can control the body weight increase of high-fat diet-induced in mice,stable glucose levels,reduce insulin resistance,reduces the accumulation of body fat and improve the lipid metabolic disorder of adipose tissue as well as inhibits macrophage infiltration.There results demonstrated that Ping tang Recipe may improve fat-induced obesity and insulin resistance by alleviating chronic low-grade inflammation of adipose tissue and fatty acid metabolism disorders to ease the adipose tissue of chronic low-grade inflammation. |
