| Objective: Detecting the expression of HBXIP and caspase9 in hepatocellular carcinoma(HCC)tissue 、pericarcinomatous tissue and normal liver tissue,and observing the apoptosis and proliferation in HCC.To investigate the effects of HBXIP and caspase 9 in the occurrence and development of HCC,as well as the correlation between the expression of HBXIP and caspase 9 and the prognosis of HCC,to provide a theoretical basis with the further diagnosis and treatment of HCC.Methods : The expression of HBXIP and caspase 9 in HCC was detected by EnVision immunohistochemical staining method in 54 cases of HCC tissue,30 cases of pericarcinomatous tissue and 15 cases of normal liver tissue.The differences of their expressions in different liver tissues were compared.The relationship between its expression and clinicopathological characteristics in HCC was analyzed.The positive expression rate of Ki 67 was detected by EnVision immunohistochemical staining method to account proliferation index(Ki67-PI),and the celler apoptosis index(AI)was detected by Tunel technique.The correlation of two proteins with the apoptosis and the proliferation were analyzed.Results:(1)The positive expression of HBXIP in HCC tissues,pericarcinomatous tissue and normal liver tissue was localized in the cytoplasm,which the positive rate were72.20%(39/54),48.30%(14/29)and 33.30%(5/15).The comparison results among three groups had significant difference(P<0.05).There was a significant difference in the positive expression rate of HBXIP betweenHCC tissue and pericarcinomatous tissue,HCC tissue and normal liver tissue(P<0.05).while the difference of the positive expression rate of HBXIP between pericarcinomatous tissue and normal liver tissue have not statistical significance(P>0.05).The positive expression of caspase 9 in HCC tissue,pericarcinomatous tissue and normal liver tissue was localized in the cytoplasm,which the positive rate were 38.90%(21/54),62.10%(18/29)and 80.00%(12/15).The comparison results among three groups had significant difference(P<0.05).There was a significant difference in the positive expression rate of HBXIP between HCC tissue and pericarcinomatous tissue,HCC tissue and normal liver tissue(P<0.05).while the difference of the positive expression rate between pericarcinomatous tissue and normal liver tissue have not statistical significance(P>0.05).(2)In HCC tissue,The positive expression rate of HBXIP in high-moderately differentiated group is lower than that in poorly differentiated group,the tumor-thrombus formation group higher than the non-tumorous embolus formation group,and the stage Ⅰ-Ⅱlower than the stage Ⅲ-Ⅳ,the differences have statistical significance(P<0.05).The positive expression rate of caspase 9 in high-moderately differentiated group is higher than that in poorly differentiated group,the metastasis group lower than the non-metastasis group,the differences have statistical significance(P<0.05).(3)There were 39 positive cases of HBXIP in HCC tissues,of which 11 cases were positive expression of caspase 9,The negative expression of HBXIP was15 cases,of which 5 cases were negative expression of caspase 9.Spearman rank correlation analysis showed that the expression of HBXIP had negative correlation with the expression of caspase 9 in HCC.(4)In HCC tissue,the average Ki 67-PI of the HBXIP protein positive expression group was 31.28±16.49,which was significantly higher thanthat of the HBXIP negative group 18.00±10.31(P<0.05).the average AI of the HBXIP positive expression group was 1.30±0.54,which was significantly lower than that of the HBXIP negative group(3.54±0.95)(P<0.05).(5)In HCC tissue,the average Ki 67-PI of the caspase 9 positive expression group was 21.19±12.63,which was significantly lower than that of the caspase 9 negative group 31.66±16.89(P<0.05).the average AI of the HBXIP positive expression group was 2.86±1.06,which was significantly lower than that of the HBXIP negative group 1.28±0.76(P<0.05).(6)The survival time of the 54 cases patients was 2-32 months.Among the 39 cases of HCC patients with HBXIP positive expression,there were 14 cases of deaths.Among the 15 cases of HCC patients with HBXIP negative expression there were 3 cases of deaths.The overall survival rate of patients with HBXIP positive expression was significantly lower(P<0.05).Among the 21 cases of HCC patients with caspase 9positive expression,there were 8 cases of deaths.Among the 33 cases of HCC patients with caspase 9 negative expression there were 9 cases of deaths(P<0.05).The comparison of the overall survival time between there two have no statistical significance(P>0.05).Conclusion:(1)The positive expression of HBXIP and caspase 9 in HCC tissues,paracancer tissues and normal liver tissues all were localized in the cytoplasm.(2)The positive expression rate of HBXIP in HCC tissues was significantly higher than that of paracancerous tissues and normal liver tissues,and was correlated with histological differentiation,TNM stage 、tumor thrombus formation and the prognosis of patients.The higher expression of HBXIP protein in HCC tissue,the more cell proliferation,the less apoptosis.The results suggest that the high HBXIP expression may play a role in the development of HCC by promoting cell proliferation and inhibiting apoptosis.It was also associated with the malignant transformation andinvasion and the clinical progress of HCC.It may serve as a tumor marker for the diagnosis and prognosis evaluation of HCC,to provide a new target for biological treatment of HCC.(3)The positive expression rate of caspase 9 in HCC tissues was significantly lower than that in paracancerous tissues and normal liver tissues,and was correlated with histological differentiation and tumor metastasis,but has no correlation with the postoperative survival time of HCC patient.The higher expression of caspase 9 protein in HCC tissue,the more cell proliferation,the less apoptosis.The results suggest that the down-regulation of caspase9 protein expression may lead to the inhibition of apoptosis,uncontrol cell proliferation,and participate in the occurrence and development of HCC.Caspase 9 was associated with the malignant transformation of hepatocytes and metastasis.It may be helpful to assess the biological behavior of HCC and to be a new target for biological treatment of HCC.(4)The expression of HBXIP and caspase 9 in HCC was negatively correlated,Which suggest that the high expression of HBXIP may lead to the down-regulation of caspase 9 protein expression,resulting in cell apoptosis and cell proliferation out of control,thus participating in the occurrence and the development of HCC.But the special mechanism needs further studied. |
