Experimental Study Of Glutathione To Promote The Healing Of Osteoporosis Bone Defect

Objective: To explore the effect of glutathione on bone healing in osteoporosis with bone defect rats,and to provide a new idea for drug therapy to promote the healing of osteoporotic fracture.Methods: 25 clean-grade SD females aged up to 6 months were randomly divided into 5 groups.The experimental group included group A,group B,group C,group D.The blank control group was Group E.There were five mice in each group.Osteoporosis model of rats in experimental group was established by bilateral ovarian resection via dorsal bilateral incision after intraperitoneal anesthesia with pentobarbital sodium.The blank control group did nothing.After 3months,the rats were examined by X-ray.Compared with the control group,the X-ray results of the experimental group showed that the brightness of bone permeability was increased and the bone structure was blurred.Then one rat was randomly selected from each group of A,B,C,D and E.These mice were removed from the femur and lumbar,which are observed the change of bone under microscope after HE staining.If the bone trabecula was sparse compared with the control group under microscope,we can determin the success of the model.A bone defect model was established with a triangular gap of about 0.5 cm in length on the lateral margin of the middle section of the right tibia after anesthesia.Then 12 rats with osteoporotic bone defect were randomly divided into group a and group b,therewere six rats in each group.These rats were placed by the same X-Ray machine for a full-body osteography.These rats with osteoporotic bone defect in group a were given a dose of glutathione at 30 mg/kg,these Rats of group b were given the same dose of saline.They were observed the healing of the bone defect by taking X-rays every two weeks.Results: There are two rats dying because of anaesthesia,one dying because of misoperation of the the experimental group 20 clean SD rats in the process of establishing osteoporosis model.In the experiment of the glutathione,four rats with osteoporotic bone defect in the experimental group were healed after 10 weeks and two rats were 12 weeks,five rats in the control group were 12 weeks and one rat was 14 weeks.The mean healing time was 10.7 weeks in experimental group and 12.3 weeks in control group.The result of two independent sample T tests was P<0.05(P=0.011).Conclusion:Glutathione can promote the repair of bone defect in osteoporotic rats with bone defect.
