Objective:By observing the effects of Dingjifumai decoction on electrocardiogram of ventricular arrhythmia rats,and on the expression of serum malondialdehyde(MDA),superoxide dismutase(SOD)content and myocardial Cx43,and to explore its curative effection and mechanism inventricular arrhythmia.Methods:Two animal models of barium chloride and aconitine were used in 80 healthy male Sprague-Dawley rats(200±20g),and Each model selected 40 healthy male SD rats were randomly divided into blank group,model group,metoprolol group,Dingjifumai decoction group,10 rats in each group.The above rats were fed normal diet and free drinking water,each group was given intragastric administration,the amount of gavage in each group was calculated according to body weight.After 2 weeks of dosing,the ECG curve of the II lead was monitored and the ECG changes were recorded by the BL-420F.Then,the aconitine and guanidinium chloride were injected into the tail vein of rats respectively.Electrocardiographic curve changes in rats.After observing the electrocardiogram,blood was collected from the abdominal aorta of rats and serum was prepared to detect the content of SOD and MDA in the serum.The heart tissue of the rats was taken,and the left ventricle and part of cardiac tissue Na~+-K~+-ATPase were selected.Immunohistochemical staining was performed on some of the rats.Immunohistochemistry(IHC)was used to measure the positive expression of Cx43 in the left ventricle of rats.Results:1.Chlorium chloride model:(1)The influence of Dingjifumai Decoction on electrocardiogram of rat model of ventricular arrhythmia induced by strontium chloride:The incidence of VP or VT in model group was 100%after intravenous cesium chloride injection,which indicated that the model of ventricular arrhythmia was successful.(1)Time of occurrence of ventricular arrhythmia:Compared with the model group,Dingjifumai decoction group and metoprolol group can significantly delay the occurrence of ventricular arrhythmia,the difference was statistically significant(P<0.05).The time of occurrence of ventricular arrhythmia was shortened in the group of Dingjifumai decoction rather than the metoprolol group(P<0.05).(2)Duration of the ventricular arrhythmia:Compared with the model group,Dingjifumai decoction group and metoprolol group can significantly reduce the duration of ventricular arrhythmia,the difference was statistically significant(P<0.05).Dingjifumai decoction group and metoprolol group had the same effect in reducing the duration of ventricular arrhythmia and there was no significant difference(P>0.05).(3)The effect of Dingjifumai decoction on the incidence of ventricular arrhythmia in rats:Compared with the model group,there was no significant difference between Dingjifumai decoction and Metoprolol group(P>0.05).The Dingjifumai Decoction group and Metoprolol group have no statistically significant difference(P>0.05).(2)Effect of Dingjifumai decoction on serum MDA and SOD in rats with ventricular arrhythmia:The serum MDA value of the model group was significantly higher and the SOD value was significantly lower than that of the blank group,and the difference was statistically significant(P<0.05).The serum MDA values of the metoprolol group and the Dingjifumai Decoction group were lower and the serum SOD value of this two group was higher than that of the model group(P<0.05).The metoprolol group and Dingjifumai Decoction group have no significant difference in serum MDA values and serum SOD values.(P>0.05).(3)Effect of Dingjifumai decoction on Na~+-K~+-ATPase in cardiac tissue of rats with ventricular arrhythmia:Compared with blank group,Na~+-K~+-ATPase value in cardiac tissue of model group was significantly decreased,and the difference was statistically significant.(P<0.05).Compared with the model group,the tissue Na~+-K~+-ATPase values of the metoprolol group and the Dingjifumai Decoction group were higher,and the difference was statistically significant(P<0.05).There was no significant difference in Na~+-K~+-ATPase activity between metoprolol group and Dingjifumai decoction group(P>0.05).(4)The effect of Dingjifumai decoction on Cx43 in cardiac tissue of rats with ventricular arrhythmia:In the blank group,more Cx43 protein expression was observed in heart tissue,and the expression of Cx43 in the model group was significantly decreased,The expression of Cx43 was observed in the metoprolol group and Dingjifumai Decoction group and was higher than that in model group.2.Aconitine model:(1)Effect of Dingjifumai Decoction on ECG in rats with ventricular arrhythmia:After aconitine injection by intravenous injection,the incidence of VP,VT and VF in the model group was 100%,which indicated that the model of ventricular arrhythmia was successful.Ventricular premature beat(VP),ventricular tachycardia(VT),and ventricular fibrillation(VF)appearance time:Compared with the model group,Dingjifumai decoction group and metoprolol group can significantly delay the occurrence of ventricular premature beats,the occurrence of ventricular tachycardia,and the occurrence of ventricular fibrillation,the difference was statistically significant(P<0.05).Dingjifumai decoction group and metoprolol group had the same effection in delaying the appearance of VP,VT and VF and have no obvious difference.(P>0.05).(2)Influence of Dingjifumai Decoction on serum MDA and SOD in rats with ventricular arrhythmia:compared with the blank group,the serum MDA level in model group was significantly increased,and the SOD value of the model group was significantly reduced,and the difference was statistically significant(P<0.05).Compared with the model group,the serum MDA values of the metoprolol group and the Dingjifumai Decoction group were lower and the serum SOD value of this two group was higher(P<0.05).There was no significant difference in serum MDA and SOD between metoprolol group and Dingjifumai Decoction group(P>0.05).(3)The effect of Dingjifumai decoction on Na~+-K~+-ATPase in cardiac tissue of rat ventricular arrhythmia:Compared with the blank group,the Na~+-K~+-ATPase value in the model group was significantly decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the tissue Na~+-K~+-ATPase values were higher in the metoprolol group and the Dingjifumai Decoction group(P<0.05);The Na~+-K~+-ATPase activity between metoprolol group and Dingjifumai Decoction group have no apparent difference(P>0.05).(P>0.05).(4)The influence of Dingjifumai decoction on Cx43 in cardiac tissue of rats with ventricular arrhythmia:The expression of brownish-yellow Cx43 protein was found in the blank group,and the expression of Cx43 was significantly reduced in the model group.The expression of Cx43 was observed in the Dingjifumai decoction and metoprolol group and was higher than that in model group.Conclusion:1.The ventricular arrhythmia rat model can be successfully prepared by intravenous injection of guanidine chloride and aconitine.2.Dingjifumai decoction can prolong the appearance time of ventricular arrhythmias induced by lanthanum chloride and shorten the duration significantly.The mechanism may be related to inward rectifier potassium channel(IK~+),which may reduce intracellular positive rate by promoting K~+outflow.Potential,reduce self-discipline and prevent arrhythmia.3.Dingjifumai Decoction can prolong the appearance time of arrhythmia such as VP,VT,VF in rats induced by aconitine,and its mechanism may be related to fast Na+channels,and may reduce the self-discipline of fast-acting cells by inhibiting Na~+influx.And prevent arrhythmia.4.Dingjifumai decoction can reduce the serum lipid peroxide content of malondialdehyde(MDA)in rats with ventricular arrhythmia,can increase the activity of superoxide dismutase(SOD),and can protect myocardial tissue Na~+-K~+-ATPase,with antioxidant free radicals,protects cardiomyocytes,thereby improving myocardial metabolism.5.Dingjifumai decoction can increase the expression of Cx43 protein in cardiac myocytes of left ventricle.
