Objections:The purpose of this experiment is to observe and compare the effect of acupuncturing the superficial fascia layer and deep median nerve of "Neiguan" on rabbits with barium chloride-induced ventricular arrhythmias.At the same time,observe and compare the effects of manual acupuncture,electroacupuncture and transcutaneous electrical stimulation of "Neiguan" on rabbits with barium chloride-induced ventricular arrhythmia model,and provide experimental basis for the clinical selection of appropriate stimulation methods.Methods:Male New Zealand rabbits were randomly divided into saline,mode,shallow needling,deep needling,manual acupuncture,electroacupuncture(EA)and transcutaneous electrical acupoint stimulation(TEAS)groups,15 rabbits in each group.All rabbits were monitored for 30 minutes by electrocardiogram(ECG)after anesthesia.The arrhythmia model was established by ear intravenous injection of Barium chloride(0.4%,1 mL/kg)at the third minute of ECG monitoring.60 seconds before molding,acupuncture needle was inserted into the superficial fascia(about 3 mm beneath the skin)or deep layer(5 to 8 mm,near the median nerve)of local PC6 tissue(upper limb)in shallow needling group and deep needling group,manipulated for a while and then retained for 10 min.The manual acupuncture group is the same as the shallow needling group.The EA group and the TEAS group were given electric stimulation for 10 min(continuous wave,4Hz,0.5mA or 2mA).At the end of ECG monitoring,the starting time and duration of arrhythmia in each group were calculated according to ECG.And histopathological changes of myocardium was observed after H.E.stain,and the immunoactivity of connexin 43(Cx43)detected by immunohistochemistry.Results:1 Interventional effect of shallow and deep acupuncture at "Neiguan" on rabbits with arrhythmia1)The average arrhythmia initiation time of the rabbits in the model group was the shortest and the longest duration of arrhythmia was(P<0.01).The average arrhythmia start time of the rabbits in the shallow needling group and the deep needling group was greater than that in the model group,the difference was statistically significant(P<0.01);the average arrhythmia duration of rabbits in the shallow-pricked group and the deep-pricked group was significantly shorter than that in the model group,and the difference was statistically significant(P<0.01).Compared with the deep needling group,the average arrhythmia start time and arrhythmia duration of the rabbits in the shallow needling group were not statistically significant(P>0.05).2)In the model group,the expression level of Cx43 in rabbit myocardium decreased seriously,and the distribution was extremely disordered.The distribution of myocardial Cx43 in the shallow needling group and the deep needling group was mild,and the expression level was significantly higher than the model group(P<0.01).3)The myocardial cells of rabbits with arrhythmia all showed pathological changes such as enhanced eosinophil staining,myocardial wave-like changes and exudation.2 The effect of different methods of stimulating "Neiguan" on rabbits with arrhythmia1)Manual acupuncture,EA,and TEAS to stimulate the "Neiguan" acupoint can delay the start time of barium chloride-induced arrhythmia(all P<0.01),and significantly shorten the duration of rabbit arrhythmia(All P<0.01).The duration of the average arrhythmia of the rabbits in the manual acupuncture group was shorter than that in the EA group and the TEAS group,but the difference was not statistically significant(P>0.05).2)The myocardial Cx43 distribution of rabbits in the manual acupuncture and TEAS group was alleviated,and the expression level was higher than that in the model group(P<0.01).The difference between the groups was not statistically significant(P>0.05).3)The myocardial cells of rabbits with arrhythmia all showed pathological changes such as enhanced eosinophil staining,myocardial wave-like changes and exudation.Conclusions:1.Both shallow needling and deep needling "Neiguan" can inhibit the arrhythmia rabbit myocardial Cx43 expression level and distribution disorder,effectively resist barium chloride-induced rabbit ventricular arrhythmia.The two are equivalent.2.Manual acupuncture,EA,and TEAS of "Neiguan" points can all improve barium chloride-induced arrhythmia model rabbits’ arrhythmia.EA may be superior to manual acupuncture and TEAS in delaying the onset time of arrhythmia.There is no statistical difference in the effect of manual acupuncture,EA and TEAS in shortening the duration of arrhythmia.Both hand acupuncture and TEAS of "Neiguan" acupoint can inhibit the degree of disordered and reduced number of Cx43 distribution in rabbit ventricular muscles induced by barium chloride.Electroacupuncture(4 Hz)has not been found to have a significant effect on inhibiting the reduction of Cx43 in rabbit ventricular muscles. |
