| Objective: To observe the differences in levels of human β-defensin-2(hBD-2),human β-defensin-3(h BD-3)and interleukin-6(IL-6)and the correlation of them in healthy subjects,patients with chronic periodontitis,patients with type 2 diabetes mellitus and patients with chronic periodontitis and type 2 diabetes mellitus by detecting the levels of them in gingival crevicular fluid of the tested individuals.To explore the possible role of human hBD-2,hBD-3 and IL-6 in chronic periodontitis and type 2 diabetes mellitus.Methods: Thirty-three with simple chronic periodontitis(CP group)were selected from Department of Stomatology of Affiliated hospital of Qingdao university,thirty-one with simple type 2 diabetes mellitus(T2DM group)and thirty-two patients with type 2 diabetes mellitus and chronic periodontitis(T2DM+CP group)were selected from Department of Endocrinology of Affiliated hospital of Qingdao university,and thirty healthy patients(H group)were selected from Center of Health Examination of Affiliated hospital of Qingdao university as control groups.The probing depth(PD),clinical attachment loss(CAL),bleeding index(BI)and gingival crevicular fluid(GCF)volume were recorded.The gingival crevicular fluid was collected,and the concentrations of human β-defensin-2 and human β-defensin-3 and interleukin-6 in gingival crevicular fluid were detected by Enzyme-linked Immunosorbent Assay(ELISA).The levels of human β-defensin-2,human β-defensin-3,interleukin-6 in gingival crevicular fluid,PD,CAL,BI and GCF volume were compared among groups.The correlation of human β-defensin-2,human β-defensin-3,interleukin-6 and periodontal parameters(PD,CAL,BI)and GCF volume were analyzed integrally and in groups.Results: The PD was highest in T2DM+CP group and higher than those in CP group,CP group were higher than T2 DM group,T2 DM group were higher than those in H group,the difference was statistically significant.The CAL in T2DM+CP group were higher than those in CP group,CP group were higher than T2 DM group and H group,the difference was statistically significant;the difference between T2 DM group and H group was not statistically significant.The BI in CP group were higher than those in H group,T2DM+CP group were higher than those in T2 DM group.The GCF volume was highest in T2DM+CP group,T2DM+CP group were higher than those in CP group,those in CP group the were higher than those in T2 DM group,T2 DM group were higher than those in H group,the difference was statistically significant.The total levels gingival crevicular fluid hBD-2 in H group were significantly higher than those in T2 DM group,T2 DM group were higher than CP group,CP group were significantly higher than those T2DM+CP group;the difference was not statistically significant between CP group and T2 DM group.The total levels gingival crevicular fluid hBD-3 in H group were significantly higher than those in T2 DM group,T2 DM group were higher than CP group,CP group were significantly higher than those T2DM+CP group;the difference was not statistically significant between CP group and T2 DM group.The total levels gingival crevicular fluid IL-6 in T2DM+CP group were significantly higher than those in CP group,CP group were higher than T2 DM group,T2 DM group were significantly higher than those in H group;the difference was not statistically significant between CP group and T2 DM group.Integrally,the level of hBD-2 was positively correlated with the level of β-defensin-3,the level of hBD-2 was negatively correlated with the level of IL-6,the level of hBD-3 was negatively correlated with the level of IL-6,the correction is significant.In groups,the level of hBD-2 was strong negatively correlated with the level of IL-6 in H group,CP group,T2 DM group and T2DM+CP group,the correction is significant.The level of hBD-3 was strong negatively correlated with the level of IL-6 in H group,CP group,T2 DM group and T2DM+CP group,the correction is significant.Conclusion: 1.hBD-2 and hBD-3 has high expression in healthy gingival crevicular fluid,and chronic periodontitis and type 2 diabetes mellitus can decrease the hBD-2 and hBD-3 concentration in gingival crevicular fluid.2.The hBD-2 and hBD-3 paly a role of protection and defense in health status,and the role under the state of chronic inflammation and type 2 diabetes mellitus can be weaken. |
