Objective This topic selected the mouse model of acute chemical liver injury induced by carbon tetrachloride,arming to reseach the protective effect of HY-12 on acute liver injury in mice and its effect on cytokines secretion.The protein and mRNA expression level of CYP2E1 and the expression level of TNF-αand IL-6 were detected to evalute the protective effect of HY-12 and its mechanism research.MethodsAll 70 kunming mice were randomly divided into normal group,model group,HY-12 high,medium and low dose group and positive control drug(glycyrrhizic group and hesperidin group).Mice with 0.5%sodium carboxymethyl cellulose(CCL4)and the corresponding drug lavage 7 days in a row.7 days after the end of the drug,in addition to the normal group of intraperitoneal injection of the same amount of peanut oil,respectively to the abdominal injection of 0.2%CCL4(0.1 ml/10 g)induced acute liver injury model in mice.Observing the effects of HY-12 in mice serum alanine aminotransferase(ALT),aspartate aminotransferase(AST)in the level of liver homogenate,and the influence of the malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione peroxidase(GSH-px)level;Liver tissue has been stained by HE.The expression of CYP2E1 in liver tissue was measured by immunohistochemistry and Western Blot.The levels of TNF-αand IL-6 in serum of mice were detected by ELISA.The expression level of CYP2E1 mRNA,TNF-αmRNA and IL-6 mRNA in the mice liver were detected by qRT-PCR.Results HY-12 could decrease ALT and AST activity in mice serum of the acute liver injury,reduce the level of MDA in liver homogenate,increase SOD and GSH-Px levels,inhibit the CYP2E1 levels in liver,and significantly reduce the degree of liver tissue-5-lesions in mice.At the same time,the middle and high dose group of HY-12 can reduce the level of TNF-αand IL-6 in liver injury mice,and reduce the degree of liver damage by inhibiting the secretion of inflammatory cytokines.Conclusion HY-12 has certain protective effect on acute liver injury in mice and its mechanism may be related to the body lipid peroxidation by inhibiting CYP2E1expression,and inhibiting the generation of inflammatory cytokines TNF-αand IL-6.
