Gout is a type of joint disease caused by the disorder of uric acid metabolism,which seriously affects the living quality of patients.The traditional Chinese medicine of Daphne Girardi could dispel wind to remove obstruction in the meridians and scattered stasis pain.At present,there are tablets and ointments of Daphne Girardi on the market,and they have obtained good clinical curative effect.But the drug loading efficiency of the ointments is lower and inconvenience to use.Therefore,the Daphne Girardi gel patch was prepared and the curative effect to treat the disease of gout was evaluated.(1)The active ingredient of daphnetin were extracted and purified,and the extraction process was optimized with single-factor and orthogonal experiment.The analysis method of active ingredients was established with the method of high performance liquid phase(HPLC).The effects of extraction times,ethanol content and material ratio on extraction amount were investigated by single factor experiment.According to the results of single factor experiment,the orthogonal experiment of three levels is carried out to optimal extraction process.The results showed that the optimal extraction process was extracted 3 times with 10 times volumes of 65% ethyl alcohol.D101 macroporous adsorption resin was used to refine the extractive,the eluent with daphnetin were collected.And the coarse powder of Daphne Girardi was obtained by dried.(2)The gel patch was prepared,and the preparation process was optimized.The results showed the optimum process was that 2.00 g absolute ethylalcohol were added,and the patch was stirred 80 s at 100 rpm.The corresponding quality standard was established to optimize the prescription of the gel patch.The dosage of 0.3% Carbomer 940 solution,sodium polyacrylate NP-700,glycerol and AlCl3 were investigated results showed that the dosages of 0.3% Carbomer 940 solution,sodium polyacrylate NP-700,glycerol and AlCl3 were 7.00 g,1.25 g,5.00 g and 25.00 mg,respectively.(3)The Daphne Girardi gel patch was prepared,and the vitro transdermal test was used to screen the skin penetration enhancer,and the results showed that the order of promoting effect of skin penetration enhancers was as follows: laurocapram>borneol>oleic acid,and the highest 24 h unit area cumulative permeability(Q24)and penetration rate(J)were obtained when 0.35 g borneol and 0.35 g laurocapram were added to the above prescription.In addition,there were no irreversible damages were observed in the pathological sections of rat skin which caused by skin penetration enhancers.(4)The curative effect of Daphne Girardi gel patch to treat gout was evaluated.The animal model of acute gouty arthritis was established by treated rats with sodium urate and colchicine was used as positive control.The swelling degree of rat ankle were measured.And the levels of TNF-α,IL-1β,SOD and MDA in the plasma of rats were determined by the corresponding kits to evaluate the anti-inflammatory antioxidant of Daphne Girardi gel patch.In addition,the curative effect of Daphne Girardi gel patch to treat gout was further confirmed by the pathological sections of rat joints and kidneys.And the results showed that the Daphne Girardi gel patch could reduce the swelling degree of the ankle joint,and significantly reduced the levels of TNF-α,IL-1β,MDA and elevate the level of SOD in rat plasma,which indicated the anti-inflammatory antioxidant effects of Daphne Girardi gel patch.In addition,Daphne Girardi gel patch could reduce the damage caused by sodium urate crystallization to the kidney,which indicated the protection effect to kidney.
