Effect Of Overexpression Of TTG-1 On Epithelial Mesenchymal Transition Induced By TGF-β1 In Lung Adenocarcinoma Cells

Objective::Through experiments,it is clear whether TTF-1 overexpression can inhibit EMT and proliferation and metastasis induced by TGF-β1 in lung adenocarcinoma cells,providing new possibility for personalized gene therapy of lung adenocarcinoma in the future.Methods:We use the lung adenocarcinoma A549 cells which were transfected with lentivirus that have been successfully constructed and verified by previous construction;The lung adenocarcinoma A549 cells were divided into three groups: TTF-1 overexpressing lung adenocarcinoma A549 cell group(experimental group),empty carrier transfected lung adenocarcinoma A549 cell group(negative control group)and untransfected lung adenocarcinoma A549 cell group(blank control group);We cultured three groups cells with a medium containing of TGF-β1,then we identify that TTF-1 overexpression of TGF-β1 induced lung adenocarcinoma cell epithelial mesenchymal transition effectsbyobserved the cell morphology,qRT-PCR and Western Blot test,and the cell migration assay,cell scratch test and cell proliferation assay.Results: The cell proliferation ability of the negative control group and the blank control group was similar,and the cell proliferation ability of the experimental group was obviously lower than that of the negative control group and the blank control group.The number of cell migration in the experimental group was less than that of the negative control group and the blank control group,and the results were statistically significant(P < 0.05).The relative distance of the scratch area in the experimental group was smaller than that of the negative control group and the blank control group.The cell migration rate of the experimental group was smaller than that in the negative control group and the blank control group,and the results were statistically significant(P < 0.05).Conclusion: 1.The overexpression of TTF-1 can inhibit TGF-β1 induced epithelial mesenchymal transition in A549 cells of lung adenocarcinoma.2.Overexpression of TTF-1 inhibits the proliferation of TGF-β1induced A549 cells in lung adenocarcinoma.3.Overexpression of TTF-1 can reduce the migration of TGF-β1 induced A549 cell migration in lung adenocarcinoma.
