The Study Of MiRNA-195 Expression In The Tissue Of Oral Squamous Cell Carcinoma And The Influence On Functions Of CAL-27 | | Posted on:2018-06-21 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y L Wang | Full Text:PDF | | GTID:2334330569986171 | Subject:Oral and clinical medicine | | Abstract/Summary: | PDF Full Text Request | | Part One The expression of mi RNA-195 in the Tissue of Oral Squamous Cell CarcinomaObjective:To detect the expression of miRNA-195 in Oral cancer tissue and normal oral mucosal tissue and to investigate whether miRNA-195 has influences on OSCC carcinogenesis and lymphatic metastasis.Methods: Select 25 cases of oral and maxillofacial squamous cell carcinoma surgical resection.The surgeries are from 2016 to 2017 in the Fourth Hospital of Hebei Medical University,among which 15 samples are from male and 10 cases are from female.Age of the patients is from 45 to 73 years old.11 cases have regional lymph node metastasis and 14 cases have non lymph node metastasis.The tumor and normal oral mucosa tissue of the same patient were taken separately.RNA was extracted from cancer tissue and normal tissue,and mi RNA-195 expression level was detected by real-time fluorescent quantitative PCR.1 The expression level of miRNA-195 in oral squamous cell carcinoma tissue and normal mucous tissue were analyzed by Statistics.The relationship between the expression of miRNA-195 and OSCC development was then analyzed.2 The expression level of miRNA-195 in oral squamous cell carcinoma tissue and normal mucous tissue were analyzed by Statistics.The relationship between the expression of miRNA-195 and OSCC lymph node metastasis was then analyzed.Results:MiRNA-195 was expressed in 25 cases of oral squamous cell carcinoma tissue and normal tissue.The result showed that miRNA-195 expression was significantly down-regulated in OSCC tissue and lymph node metastatic group compared to normal mucous tissue and non-metastatic OSCC group.These datas suggested that miRNA-195 expression is negatively correlated with OSCC development and Lymph node metastasis.Conclusion:1 The mi RNA-195 expression was significantly down-regulated in OSCC tissue compared to normal mucous tissue.2 The miRNA-195 expression was significantly down-regulated in lymph node metastatic OSCC compared to non-metastatic OSCC.Part two:Study of the effect of miRNA-195 on the biological function of CAL-27 cellsObjective: To investigate the influences of miRNA-195 on proliferation and migration of CAL-27 and to explore the role of miRNA-195 in carcinogenesis and lymphatic metastasis of OSCC.Methods:1 The recovery,cultivation,passage and frozen storage of OSCC-15CAL-27 cells.2 Synthetic miRNA-195 mimics or miRNA-195 NC was immediately transfected into OSCC-15 seperately.3 The proliferation of CAL-27 was detected by MTS method and single-colony formation assay.4 The migration ability of CAL-27 was measured by transwell migration assay.Results: Compared with control group,the proliferation and migration of cell were significantly decreased in the cells transfected with miRNA-195.Conclusions: MiRNA-195 is a tumor suppressor of OSCC,which could inhibit proliferation and migration of CAL-27 cells.Mi RNA-195 might be a new potential target for OSCC therapy. | | Keywords/Search Tags: | OSCC, MiRNA-195, Q RT-PCR, CAL-27 cell, Metastasis Proliferation, Migration | PDF Full Text Request | Related items |
