Effects Of Different Concentrations Of Pomegranate Peel Polyphenol Ointment On Histopathological Morphology Of Rabbit Ear Acne Model

Object:This study used the different concentrations of pomegranate peel polyphenols ointment preparation and used in rabbit ear acne model,the method of observation under different concentrations of pomegranate peel polyphenols ointment histopathologic morphology of rabbit ear acne model.Methods:Will buy pomegranate peel polyphenols powder,according to the matrix compatibility proportion of ointment,configured to 3%,9%and 18%respectively of pomegranate peel polyphenols ointment concentration.And its shape,PH,stability,skin irritation reaction to do evaluation.Will be 42 male and female half weight between 2.0 to 2.5 kg of white rats were randomly divided into 7 groups,respectively,the blank group,model group,the matrix group,the positive group,low dose group,middle dose group,high dose group.Will group 7 rabbits except the blank group,respectively in the outer white rabbit left and right external auditory canal and place is about 2.0 cm x 2.0 cm range inside and outside is about 0.5 mL,2%coal tar coating solution 1/d(coal tar)mixture 2%of coal tar with 95%alcohol solution,for 2 weeks to make the animal model of acne.Empty since given normal saline group and model group,daily from the rabbit ears with 0.1 ml of normal saline.About 3 groups for matrix,daily and ear Angle place with 0.1 g of matrix.Low dose,middle dose and high dose group,and every day around diversification in besmear respectively 0.1 g of 3%,9%and 18%of pomegranate peel polyphenols ointment.Positive group,and every day in the left and right ear Angle is changed with 0.025%vitamin A acid cream o.1 g.Apply twice a day for 3 weeks intervention,dosing parts are made of rabbit ear acne model.In the last 24 h after dosing to 3 mm punch production coated bolts specimens(full-thickness skin),with 10%formalin fixed,paraffin embedding,sectioning,HE staining,under a microscope to observe the histopathological changes,and micrography.Results:1.The low dose group,middle dose group,high dose ointments three concentrations of pomegranate peel polyphenols color into a grey,properties of milk cream,daub on the skin can feel the smooth texture,no particles,with good malleable.Three different concentrations of cream there is no obvious difference in character.Three different concentrations of pomegranate peel polyphenols ointment PH value is neutral.and good stability.Low,medium and high dose of pomegranate peel polyphenols ointment in 3 weeks no obvious skin irritation reaction.2.Normal group compared with model group in histopathology,the level of the degree of inflammatory,and degree of cuticles,etc were showed significant difference(P<0.05;P<0.01),and base on the above indexes influence was not significant(P>0.05).low dose group and high dose group have certain effects on the above indexes(P<0.05),but not as good as the influence of the dosage group and positive medicine group significantly(P<0.05,P<0.01).Conclusion:l.High,medium and low dose of pomegranate peel polyphenols ointment properties stable.3 weeks of rabbit skin irritation reaction is not obvious.2.The dosage group and positive medicine group on the Angle of rabbit ear acne model and the most significant anti-inflammatory effect...
