Objectives:In this paper,the pre-treatment process of salvia miltiorrhiza preparation process were screened and optimized,and the parameters were determined in the process of extraction,concentration and spray drying of salvia miltiorrhiza,so that they could provide a reference for specifications in the productive process of salvia miltiorrhiza.This topic has also studied the physical properties of the extraction,enrichment and the products of the spray drying,and has been built out of the physical parameters of relevant mathematical models which were used to perfect the quality evaluation system of salvia miltiorrhiza.To expand physical parameters research significance,thus for the study of the physical parameters could be more widely used to provide theoretical foundation and data base.Methods:(1)The HPLC method was used to establish the method for content determination of the fat-soluble component Tanshinone ?A and the water-soluble component Salvianolic acid B in Salvia miltiorrhiza Bunge,and make a study on methodology.(2)The HPLC method was used to investigate the content of the index components in salvia miltiorrhiza under different ways of extraction and enrichment,and compares the differences of them.Using the SAS data analysis software for data analysis of variance.(3)The density of salvia miltiorrhiza extract and concentrate were determined by float type densimeter.The brix degree of extract and concentrate were investigated with Abbe refractometer.By rotating viscosity meter to determine the apparent viscosity of salvia miltiorrhiza extract.The thermal conductivity instrument was used to analysis thermal conductivity of salvia miltiorrhiza extract.Using specific heat meter to measure the specific heat of different concentration of salvia miltiorrhiza extract and concentrate.(4)By using Excel and Origin drawing software to extract the experimental data of physical parameters monadic linear regression analysis.Using 1stopt data analysis software for some physical parameters for binary nonlinear fitting and binary linear regression analysis.Using MATLAB simulation software to simulate the response surface of the physical parameters.(5)The single factor analysis and Box-Behnken Design response surface method were used to investigate the change of the yield and water content in the spray drying process of salvia miltiorrhiza water extract and alcohol extract,and compare the powder wall sticking rate differences.(6)Halogen rapid moisture tester was used to determination the water content of spray drying powder which were from salvia miltiorrhiza water extract and alcohol extract of spray drying powder.The morphology and particle size of spray dried product were investigated by means of environmental scanning electron microscopy(SEM).Using the rest Angle tester to determine the powder Angle of repose.Manual vibration wave method was used to determination of powder bulk density and compaction density,and calculate the degree of compression.Examine the hygroscopicity of salvia miltiorrhiza powder under different humidity environmen.Examine the hygroscopicity of salvia miltiorrhiza powder under different humidity environment.Results:(1)The method for the determination of Tanshinone ?A and Salvianolic acid B in Salvia miltiorrhiza Bunge were established by HPLC.The regression equation of Tanshinone ?A was:Y = 6462576 X-11003(r = 0.9999)and the linear range was 0.032?0.640 ?g.The linear regression equation of the obtained Salvianolic acid B was:Y = 1166190 X + 21684(r=0.9999),and the linear range was 0.700?11.200?g.(2)Using high performance liquid chromatography method to investigate the different extraction solvents extraction efficiency,determine the 60%ethanol extract and water reflux extraction way of major component extraction efficiency and cost savings.The change of the concentration of the extract in the concentration of the extract was investigated.It was found that in the process of reduced pressure concentration high transfer rate and concentration index linear component content changed little.(3)The physical parameters of salvia miltiorrhiza extract were determined and related mathematical models were constructed.Established the equations of crude drug concentration-density and concentration-brix degree respectively,and determine the brix degree method to determine the concentration of the characterization.To establish the relationship between viscosity of salvia miltiorrhiza extracts and its concentration was ?=AeBC,the relationship model of viscosity and temperature was ?= AeB/T and the relationship between the viscosity and concentration,temperature was ?=cex(Ea/RT+dC+fC2).Determination of salvia miltiorrhiza extract relationship between the thermal conductivity and the temperature:? = a-bT,the relationship between the thermal conductivity and the concentration was:??a-bC,and the model of the thermal conductivity,temperature and concentration was:?= a-bC-cT-dCT.The heat capacity of the investigation and the salvia miltiorrhiza extract relation with concentration model was:cp =a-bC.(4)According the datas of physical properties to calculate the thermal diffusivity and the Prandtl number and analysis of their rule.Renault numeral and Nusselt numeral were derived,and analyze the significance of the application.(5)According to the results of response surface method optimization,get the water extraction liquid of salvia miltiorrhiza in spray drying process was:the material density was 1.15 g/cm3,the inlet temperature was 200 ?,the feed rate was 40 r/min.On the other hand the salvia miltiorrhiza alcohol extraction liquid spray drying process is:material liquid density of 1.13 g/cm3,the inlet air temperature of 160?,the feed rate of 45 r/min.Examine and compare the condition found that the wall sticking condition of salvia miltiorrhiza alcohol extraction spray powder was more serious than the water extraction.(6)Comparison of salvia miltiorrhiza water extraction liquid spray drying powder and alcohol extraction liquid spray drying powder of various physical parameters.The results showed that the water content of salvia miltiorrhiza water extraction liquid spray drying powder(5.82%)is less than alcohol extract powder(6.52%),and particle size were smaller and less liquid,but the moisture absorption under different humidity environment were larger.Conclusions:The method for the determination of Tanshinone ?A and Salvianolic acid B were exclusive and reproducible.They could be used for the determination and quality control of the essential components in Salvia miltiorrhiza.The method of determination the physical properties used in the extraction,concentration and spray drying process of salvia miltiorrhizaa was effective,simple and accurate.The physical models were well fitted and could be used to characterize the change of the physical properties.The optimal spray drying process was fast and efficient,and the products' quality were better.The characteristics parameters of these products were accurate and effective.They have guiding significance to analyze the physical properties of traditional Chinese medicine pharmaceutical,to select or design the equipment of Chinese medicine production and they were suitable for the production process control,etc.
