Objective:To explore the effects of electroacupuncture at Guanyuan(CV4)or Sanyinjiao(SP6)acupoint in nine-month-old female mice with senescence-accelerated mouse prone 8(SAMP8),for providing the new thinking and the objective scicence evidence for treatment and prevention Alzheimer’s Disease(AD).Based on the results,to discuss the mechanism of improving the learning and memory ability by EA-Guanyuan or EA-Sanyinjiao in nine-month-old SAMP8 female mice,to research the correlation of between hypothalamus-pituitary-ovary axis(HPO)and AD,meanwhile to analyze the relative acupoints specificity,to providing the experimental evidence for choosing acupoint in clinic.Methods:18 Nine-month old female mice with senescence-accelerated mouse prone 8(SAMP8)were divided into three groups:the model control group,tihe EA-Guanyuan treatment group and the EA-Sanyinjiao treatment group,six in each group.Meanwhile,9-month old female mice with senescence-accelerated mouse resistance l(SAMRl)female mice were set as the blank control group.The two treatment groups were given the same pattern of EA stimulation.To oberve the general condition of the mice in each group,such as hair condition,defecation state and mental state;The gonadotropin-releasing hormone(GnRH),luteinizing hormone(LH),follicle-stimulating hormone(FSH),E2 levels of HPO axis as well as the β-amyloid precursor protein(P-APP),Aβ1-42 levels of cerebral cortex were detected and recorded using enzyme-linked immunosorbent assay(ELISA)method,the learning-memory ability was assessed by the Morris Water Maze(MWM)test.SPSS statistical software was used for analysis.Results:1.Normal conditions:1)Normal condition:The blank control group behaved active responds and high spirit.The model controlgroup were characterized by less active,unresponsive,low enthusiasm.Compared with the model group,two EA groups performed better,EA-Sanyinjiao group was better.2)Fur state:The blank control group with white and shinning coat.While,there were different degrees of hair removal and dim yellow coat color in the model control group.After EA stimulation treatment,fur color and hair removal conditions were improved.3)Stool texture:The blank control group were normal,the model control group were dry stool.Two EA groups had a soft stool,EA-Guanyuan group was better.2.The MWM test results1)Compared with the blank control group,the model control group spended escape latency period prolonged every day,the model control group had weak learning and memory ability(p<0.01).2)Compared with the model group,Both of two EA groups had shortened escape latency period,the response latency in the EA-Sanyinjiao treatment group was found to have statistical significance every day(P<0.05 or P<0.01),while the EA-Guanyuan treatment group only had significantly decreased on the fifth day.Contrast with the two EA groups,there was statistical significance on the fifth day(p<0.05),other four days were not obvious(p>0.05).3.Effects of EA-Guanyuan or EA-Sanyinjiao acupoint on HPO axis1)Compared with the blank control group,E2 level had significantly decreased(P<0.01),the levels of GnRH and LH.FSH levels increased obviously in the model control group(P<0.01;P<0.01;P<0.01).2)Compared with the model group,Ovarian E2 level of the Two EA groups were increased in different degree,EA-Sanyinjiao group had obviously boosted(P<0.01),EA-Guanyuan group had boosted(P<0.05),GnRH and LH、FSH levels were declined in different degree,what is more,the EA-Sanyinjiao group had significant difference(P<0.01;P<0.05;P<0.01),the EA-Guanyuan group had difference(P<0.05;P<0.05;P<0.05);The difference between two EA groups was not obvious.4.Effects of EA-Guanyuan or EA-Sanyinjiao acupoint on β-APP、Aβ1-42 levels in the cerebral cortex1)Compared with the blank control group,β-APP and Aβ1-42 levels increased obviously in the model control group(P<0.01;P<0.01).2)Compared with the model group,β-APP and Aβ1-42 levels decreased obviously in the EA-Sanyinjiao group(P<0.05,p<0.05),through the levels of p-APP and Ap1-42 had decreased trendency in EA-Guanyuan group,but the difference was not obvious(p>0.05).Contrast with the two EA groups,the difference of Aβ1-42 was obvious(P<0.05);although the decreased trendency of β-APP had difference between the two EA groups,but there was not statistical significance(P>0.05).Conclusion:1 Both of electroacupuncture of Guanyuan(CV4)and Sanyinjiao(SP6)could have an effective impact on the disordered function of HPO axis,effectively rising estrogen levels in SAMP8 female mice and declining the hypersecretion of hypothalamus and pituitary hormone.2 Electroacupuncture can restrain the expression of β-APP and Aβ1-42,improve the learning and memory ability in AD model female mice,the mechanisam maybe through adjusting the hpo axis function and related hornmone secretion.3 Sanyinjiao acupoint maybe is superior on preventing and treating AD disease than Guanyuan acupoint.
