| migraine is a common disease of the nervous system.It is closely related to a variety of diseases such as cerebral infarction and depression,seriously affecting the health of human beings,because of its high incidence,repeated attack,severe pain in itcal time and protraction,causing infinite pain to the patients.Consequently,migraine prevention is an important topic of the medical research.The treatment of migraine according to syndrome differentiation based on the overall concept of traditional Chinese medicine,has its own advantages and characteristics.However,the relative weakness of fundamental research limits the play of the advantages--the prevention and treatment of migraine with Chinese medicine,so we should combine the traditional Chinese medicine theory with modern medical frontier,further consider its deep mechanism,provide new evidence supporting for it and promote its better development and more widely spread.Objective:1.To explore the expression of PPARs subtypes in the trigeminal cervical complex(TCC)at different time points in migraine rats and analyze the relative peak time of PPARs expression.2.To study the effect of Tianshu Capsule on the expression of PPARs subtypes in the trigeminal cervical complex of migraine rats and compare the changes of CGRP in the blood of rats treated with Tianshu Capsule and PPARs agonists,clarifying the mechanism of Tianshu Capsule in the treatment of migraine.MethodsPart I.Expression of PPARs subtypes in the trigeminal cervical complex of migraine rats at different time points.1.24 SD rats were divided into model group and blank group.The rats of blank group were fed normally and the rats of model group were treated with nitroglycerin by subcutaneous injection.2.Continuously recording for 2 hours after modeling and observing the behavior changes of rats.3.After modeling for 2 hours,six rats were taken from each of the blank group and the model group.The brain tissues were collected after anesthesia and decapitation respectively.After modeling for 4 hours,the brain tissues were collected using same method.4.The expression of PPARs subtypes in TCC was observed by immunohistochemistry.Part Ⅱ.Study on the regulatory effect of Tianshu Capsule on the expression of PPARs1.32 rats were divided into 4 groups,including blank group(normal feeding,no treatment),model group(modeling after 7-day intragastric administration with normal saline),Tianshu Capsule group(modeling after 7-day intragastric administration with Tianshu Capsule aqueous extract),GW0742 group(intraperitoneal injection with GW0742 after 7-day intragastric administration with normal saline,and modeling after intraperitoneal injection for half an hour).2.Continuously recording for 2 hours after modeling and observing the behavior changes of rats.3.After modeling for 4 hours,the blood was collected from femoral artery;the brain tissues were collected after decapitation.4.The CGRP content in plasma was measured using ELISA.The determination method of PPARs subtypes was same as Part Ⅰ.Results:1.The expression trend of PPAR a in TCC of migraine rats is descending first and then ascending;the expression trend of PPAR β in them is keeping increasing in 4 hours;the expression trend of PPARγ in them is ascending first and then return to the normal level.2.Both Tianshu capsule and GW0742 can reduce the migraine behavior score of the migraine rats,and then they can be used to treat the migraine.3.Tianshu capsule can raise the expression of PPAR a and PPAR β in TCC,but it have no effect on the expression of PPAR y;GW0742 can raise the expression of PPAR 0,restrain the expression of PPAR α,and have no effect on the expression of PPAR γ.4.Both Tianshu capsule and GW0742 can descend the level of CGRP in blood plasma of the migraine rats in model group.Conclusions:1.Tianshu capsule have a definite positive effect on prevention the migraine.2.Tianshu capsule can prevent the migraine by activating the expression of PPAR a and PPARB in the trigeminal nerve cervical composites and descend the level of CGRP in blood plasma.3.Tianshu capsule has the same effect as PPAR β agonist,and it can prevent the migraine by activating the expression of PPAR β,but its action effect is different from PPAR 0 agonist.4.Maybe PPARs is a new target spot for preventing the migraine with Chinese medicine in the future. |
