Objective To study relationship betwecn P12A Single nucleotide polymorphism in peroxisome proliferator-activated receptors gamma2 (PPAR gamma2) and type 2 diabetes and obesity. Methods According to WHO criteria for diagnosis of diabetes mellitus,the control group consisted of 55 subjects,type 2 diabetes mellitus 97 subjects.According to BMI in type 2 diabetes mellitus,normal weight group(54) and overweight/obese group(43) were included in this study。These subjects were evaluated for the Pro12Ala single nucleotide poly-morphism in the PPAR gamma2 using Taqman probe in real-time Q-PCR.. Results A allele frequencies of Pro12Ala mutation of peroxisome proliferator-activated receptors gamma2(PPAR gamma2) were 5.5% and 7.7% in control group and type 2 Diabetes Mellitus.There were no differences in genotype and allele frequencies between those two groups(P>0.05)。After adjustment for age,sex,BMI,there were detected evidence for an interactin effect between the Pro 12Ala single nucleotide polymorphism and type 2 diabetes using logistic regression , OR=1.237,95%confidence interval 0.363~4.213. A allele frequencies of Pro12Ala mutation of peroxisome proliferator-activated receptors gamma2(PPAR) were 9.3%,6.5% in obese group and non-obese group with 2 cliabetes. There were no differences in genotype and allele frequencies between those two groups(P>0.05 ). After adjustment for course of diseases,there were no relationship between the Pro12Ala single nucleotide polymorphism of peroxisome proliferators-activated receptor gamma2 and with obese diabetes using logistic regression, OR=1. 917,95%confidence interval 0.586~6.273.The Pro12Ala polymorphism of peroxisome proliferators-activated receptor gamma2 in diabetes patients were associated with increased BMI,waist circumference and waist to hip ratio (P<0.05).Insulin resistant index of the subjects which the allele is A is lower than that the subjects which the allele is P ,there were no relationship between the Pro12Ala variant of PPAR gamma2 and insulin resistant(P=0.053).Conclusion The Pro12Ala single nucleotide polymorphism of peroxisome proliferator-activated receptors gamma2 was not associated with either diabetes or obesity with diabetes. The Pro12Ala single nucleotide polymorphism of peroxisome proliferator-activ ated receptors gamma2 could be association with abdominal obesity in type2 diabetes .
