Purpose: Basing on Adipo R1/AMPK signal pathway,it studied the compatibility of Astragalus and Puerariaelobatae Radix effected on liver glucose-lipid metabolism and inflammation in diabetic rats.It explored the mechanism of Astragalus,Puerariaelobatae Radix compatibility of glucose-lipid metabolism and inflammatory reaction in diabetic rats.The interactions of the compatibility of Astragalus and Puerariaelobatae Radix regulating the glucose-lipid metabolism and inflammation in diabetic rats is analyzed.All of those make foundation to use reasonable drugs and develop new drugs for diabetes in clinic.Material and method: The 86 male SD SPF rats,according to blood glucose factors were randomly divided into 6 groups:(1)11 in normal group,(2)15 in model group,(3)15 in positive control group,(4)15 in Astragalus Group,(5)15 in Puerariaelobatae Radix Group,(6)15 in Huangqi Gegen Decoction group.Except the normal group,the rats were given a one-time intraperitoneal injection of streptozotocin(STZ)46 mg·kg-1 to induced the diabetic model.Given 10 m L·kg-1 distilled water to normal group,10 m L·kg-1 distilled water to model group,1.47g·kg-1 Jinqi Jiangtang tablet to positive control group,2.7g·kg-1 Astragalus to Astragalus Group,1.35g·kg-1 Puerariaelobatae Radix to Puerariaelobatae Radix group and 4.05g·kg-1 Huangqi Gegen Decoction to Huangqi Gegen Decoction group to receive gavage,each group was given by 30 days continuously.After 30 days successive medication,measuring fasting blood glucose by blood glucose meter,serum cholesterol(CHO)and triglyceride(TG)by biochemical analyzer and the contents of insulin(Ins),insulin receptor(Ins R)and tumor necrosis factor α(TNF-α)in rat liver tissue by enzyme-linked immunosorbent assay(ELISA).By using quantitative real-time PCR(QPCR method)to detect adiponectin receptors-1(Adipo R1),AMP-activated protein kinase(AMPK),glucose transporter 4(GLUT-4),p38 mitogen-activated protein kinase(p38 MAPK),peroxisome proliferator-activated receptor α(PPARα)m RNA expression in rat liver tissues.The experimental datas were analyzed by SPSS 17.0.Results: 1.Effect on blood glucose and blood lipid of diabetic rats(1)Effect on the blood glucose of diabetic rats: the blood glucose of rats in model group was significantly higher than the other groups.The blood glucose of rats in Astragalus group,Puerariaelobatae Radix group,and Huangqi Gegen Decoction group were significantly reduced.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).(2)Effect on the blood fat of diabetic rats(1)Effect on CHO of diabetic rats: The serum level of CHO in model group was significantly increased,and the serum CHO level was significantly lower in the group of Astragalus group,Puerariaelobatae Radix group,and Huangqi Gegen Decoction group.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).(2)Effect on TG of diabetic rats: The serum TG of rats in model group was significantly increased,and the serum TG of rats in Astragalus group,Puerariaelobatae Radix group and Huangqi Gegen Decoction group were significantly lower.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had statistical significance(P<0.05).The compatibility of Astragalus and Puerariaelobatae Radix worked better than single Astragalus or Puerariaelobatae Radix.2.Effect on Ins,Ins R,TNF-α level in liver of diabetic rats(1)Effect on Ins level in liver of diabetic rats: The liver Ins level of rats in model group was significantly increased,and the liver Ins level of rats in Astragalus group and Huangqi Gegen Decoction group were significantly lower.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).(2)Effect on Ins R level in liver of diabetic rats: The liver Ins R level of rats in model group was significantly increased,and the liver Ins R level of rats in Astragalus group,Puerariaelobatae Radix group and Huangqi Gegen Decoction group were significantly lower.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had statistical significance(P<0.05).Astragalus worked better than the compatibility of Astragalus and Puerariaelobatae Radix.(3)Effect on TNF-α level in liver of diabetic rats: The liver TNF-α level of rats in model group was significantly increased,and the liver TNF-α level of rats in Astragalus group,and Huangqi Gegen Decoction group was significantly lower.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had statistical significance(P<0.05).Astragalus worked better than the compatibility of Astragalus and Puerariaelobatae Radix.3.Effect on Adipo R1,AMPK,GLUT-4 m RNA expression level of diabetic rats liver(1)Effect on Adipo R1 m RNA expression level of diabetic rats liver: The liver Adipo R1 m RNA expression level of rats in model group was significantly lower,the liver Adipo R1 m RNA expression level of rats in Astragalus group,Puerariaelobatae Radix group and Huangqi Gegen Decoction group were increased significantly,the difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).(2)Effect on AMPK m RNA expression level of diabetic rats liver: The liver AMPK m RNA expression level of rats in model group was significantly lower,the AMPK m RNA expression level of rats in Astragalusi group and Huangqi Gegen Decoction group were increased significantly.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).(3)Effect on GLUT-4 m RNA expression level of diabetic rats liver: the liver GLUT-4 m RNA expression level of rats in model group was significantly lower,the GLUT-4 m RNA expression level of rats in Astragalus group,Puerariaelobatae Radix group and Huangqi Gegen Decoction group were increased significantly.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).4.Effect on p38 MAPK,PPARα m RNA expression level on diabetic rats liver(1)Effect on p38 MAPK m RNA expression level on diabetic rats liver: the liver p38 MAPK m RNA expression level of rats in model group was significantly higher,the liver p38 MAPK m RNA expression level of rats in Astragalus group,Puerariaelobatae Radix group and Huangqi Gegen Decoction group was decreased significantly.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had no statistical significance(P>0.05).(2)Effect on PPARα m RNA expression level on diabetic rats liver: the liver PPARα m RNA expression level of rats in model group was significantly lower,the liver PPARα m RNA expression level of rats in Astragalus group,Puerariaelobatae Radix group and Huangqi Gegen Decoction group was increased significantly.The difference was statistically significant(P<0.05).The interaction of Astragalus and Puerariaelobatae Radix had statistical significance(P<0.05).Astragalus worked better than the compatibility of Astragalus and Puerariaelobatae Radix.Conclusion: 1.Astragalus,Puerariaelobatae Radix and Astragalus-Puerariaelobatae Radix compatibility(Huangqi Gegen Decoction)can reduce the blood glucose of diabetic rats.Huangqi Gegen Decoction does not show Astragalus,and Puerariaelobatae Radix synergistic interaction,which shows that it is not simply overlay relationship of the prescription in regulating blood glucose.2.Astragalus,Puerariaelobatae Radix and Astragalus-Puerariaelobatae Radix compatibility(Huangqi Gegen Decoction)can reduce the blood lipid of diabetic rats.In reducing CHO,Astragalus and Puerariaelobatae Radix has no interaction.In reducing TG,Astragalus and Puerariaelobatae Radix has the interaction.The compatibility of Astragalus and Puerariaelobatae Radix worked better than Astragalus or Puerariaelobatae Radix,which shows the selectivity of the prescription worked on the body of synergistic or antagonistic effects.3.Regulation blood sugar of Astragalus,Puerariaelobatae Radix and AstragalusPuerariaelobatae Radix compatibility(Huangqi Gegen Decoction)may be related to the regulation of liver Ins and Ins R levels.Astragalus,Puerariaelobatae Radix and Astragalus-Puerariaelobatae Radix compatibility in regulating glucose-lipid metabolism and inflammatory reaction may be associated with the Adipo R1/AMPK signal pathway.
