| Purpose:First,it is preferable to optimize the ethanol extraction process of total phenolic acid in clove leaves and prepare the oral solution.The second is to establish a method for the determination of three representative components in clove leaves oral liquid;Through the pharmacological and toxicological studies of clove leaves oral liquid,to explore its anti-bacterial anti-inflammatory treatment and drug safety.Method : The optimum amount of total phenolic acid in clove leaves was determined by orthogonal test.The optimum extraction conditions were as follows: water extraction rate,boiling time and boiling time were selected as the evaluation indexes.Water extraction process,the alcohol concentration,alcohol precipitation time and alcohol precipitation temperature were selected as the factors.The orthogonal experiment was used to optimize the ethanol precipitation process of clove leaf extract.The relative correction factor of protocatechuic acid and protocatechuic aldehyde and caffeic acid was determined by HPLC with the method of protocatechuic acid as the reference substance and external standard method in the oral liquid of clove leaves.The amount of catechins and caffeic acids in clove leaves oral solution was determined by external standard method,and the difference between them was compared with that calculated by using relative correction factor.At the end of this study,the anti-inflammatory and anti-inflammatory effects of clove lipstick oral liquid were investigated by p-xylene-induced mouse ear swelling and carrageenan-induced rat swelling.The acute toxicity test was used to investigate its safety.Results: 1.The optimum extraction process was 10 times the amount of water,boiling 3 times,each time 1h。 2.the best alcohol precipitation process for the alcohol content of 70%,temperature60 ℃,standing 12 h.3.The relative correction factor was reproducible.There was no significant difference between the calculated values of the relative correction factors and the external standard method.4.Compared with the blank control group,P <0.05 had significant effect on the high dose group and the positive control group.No tolerance to animals at maximum tolerated dose.Conclusion: 1.The process is simple and stable,and it can be used as the extraction process of clove leaves.2.It is applicable and feasible in the determination of three phenolic acids in oral administration of clove leaves,which can reduce the use of other reference substances.3.Clove leaves oral liquid anti-bacterial anti-inflammatory effect of the exact,non-toxic side effects. |
