1.Research backgroundDepression is a complex emotional disorder.The patients mainly manifested as significant and lasting depressed mood,the duration is about two weeks or more than two weeks,and often have the corresponding abnormal thinking and behavior occurs,the incidence of the high rate,high disability rate and the rate of Dutch act has become the major cause of disability in patients with depression in developed countries.In China,the incidence of depression is also increasing year by year.According to statistics,the number of diseases has reached nearly thirty million people,and is in the forefront of the world's major diseases.Studies have shown that treatment of depressive symptoms in patients with heart disease can not only improve depressive symptoms.Some studies have shown that the severity of depression is positively correlated with cardiovascular events and is an independent risk factor for cardiovascular disease,but it is an independent risk factor for cardiovascular disease,but it has been found to be an independent risk factor for cardiovascular disease.There is no clear conclusion of the pathogenesis.But also affect the occurrence and prognosis of cardiovascular diseases.At present,the diagnosis and treatment of depression has not yet found a specific method.Long term use of Western medicine gives the patient a different degree of side effects.And acupuncture treatment of depression patients with sleep,anxiety,cognitive impairment,diurnal changes and so on have a more obvious improvement.Acupuncture is simple in operation,convenient in treatment,small in side effects compared with western medicine,low in price and fast in effect.It provides a new way of thinking for clinical treatment of depression.2.ObjectiveExperiments show that whether electroacupuncture antidepressant treatment will affect the plasma of rats with chronic stress depression in 6-keto-PGF1,TXB2,alpha P-selectin,and compared with the effect of drug treatment,so as to further clarify the effect of depression related to the pathophysiology of cardiovascular disease mechanism.3.Methods1.Animal groupsChoose a healthy adult male SD 80 rats,fed for 1 weeks,by the open field test of rats were screened out,of which the horizontal and vertical motion number was less than 30 or greater than 120 times removed,only 32 remaining rats were randomly divided into two groups,namely control group,model control group,drug control group and treatment group,8 rats in each group.Then,chronic stress stimulation and different interventions were initiated.In the blank group,the rats were still kept in a cage free mode,while the rest rats in each group adopted solitary feeding mode.2.Modeling methodAccording to the literature method,chronic stress model was adopted,and all the rats in the model rats were randomly assigned to 21 days and 7 different stress stimuli.Including cold water swimming 5min,day and night upside down 24h,fasting 24h,water prohibition 24h,tail 3min,wet environment 24h,binding 3H and other stimuli.One kind of stimulus was randomly assigned daily,with an average of 3 times per stimulus.3.Processing methodThe blank group:under the normal feeding condition of 21 days,the non-soliton mode was adopted,and the stress stimulation was not given during the experiment.Model control group:given unpredictable stress stimulation for 21 days,made a model with chronic unpredictable stress depression,and in 21 days,it was equal to the electroacupuncture group 'restraint intervention,each intervention time,length and intensity were the same.The drug control group:chronic stress with solitary and daily Prozac with distilled water dispensed according to the ratio of 1mg/1ml,according to 2mg/kg weight,intragastric administration,rats after 1h stress.Once a day,a total of 21 days.Treatment group:chronic stress with solitary and "Baihui" and "Yintang" point flat thorn needle stimulation,acupuncture treatment for 21 days,a day after the 1H stress model of electroacupuncture.4.Detection indexes and methodsThe open field test(Open-field Test),sugar consumption experiment(Sucrose Preference Test)and broken blood,plasma levels of 6-keto-PGF1 alpha,TXB2,P-selectin content by ELISA method,the content was determined by HPLC in hippocampus and cortex tissues of 5-HT,DA,NE.4.Result1.Behavioral changesBefore the experiment,there was no statistical difference in the behavior of rats in each group(P>0.05).After modeling,the rats relative sucrose consumption,horizontal movement and vertical movement were significantly different(P<0.05),the rats in the model group of horizontal movement and vertical movement number were significantly lower than that of the control group(P<0.05),EA group movement number was significantly higher than the model group(P<0.05).2.Changes of the content of 5-HT,DA and NE in 2.hippocampusThe contents of 5-HT,DA and NE in hippocampal tissues of each group were significantly different(P<0.05),and the contents of 5-HT,DA and NE in model group were significantly lower than those in blank group(P<0.05),and the contents of 5-HT,DA and NE in drug group and EA group were significantly higher than those in model group(5-HT<0.05).3.Changes of the content of 6-keto-PGF1 alpha,TXB2 and P-selectin in 3.plasmaThe plasma levels of 6-keto-PGF1 alpha,TXB2,P-selectin content had significant difference(P<0.05),the 6-keto-PGF1 drug group and EA group in rat plasma alpha were significantly higher than those in the model group(P<0.05),TXB2 and P-selectin content were significantly lower than the model group(P<0.05).5.ConclusionElectroacupuneture treatment with antidepressant effect,can effectively improve the behavior and hippocampus of chronic stress depression rats in 5-HT,DA and NE levels,and the content of the 6-keto-PGF1 alpha in plasma increased TXB2 and decreased the content of P-selectin,further suggested that depression may cause platelet activation through affecting 6-keto-PGF1 alpha/TXB2 and P-selectin thus,influence the occurrence and prognosis of cardiovascular disease.
