| In the middle of the 19th century,the term lupus erythemateux(LE)appeared,describing cases of ulcerative lesions with facial erosion.With the development of modern medicine,LE was constantly aware of the research.LE includes system type and skin type and system type has occurred all over the world.Differences between different countries and regions are more obvious.The prevalence of systemic lupus erythematosus is high in China.The prevalence of systemic lupus erythematosus is high in China and it is common in women of childbearing age.The incidence is increasing year by year.The patient has caused great health and property losses,while the lack of epidemiological data of cutaneous lupus erythematosus in China.Lupus erythematosus is an autoimmune disease and often accompanied by the involvement of multiple organs,so the disease will enable the patient to produce antibodies against his own cell.The clinical manifestations are complex,difficult to cure,and the condition is prone to repeated and poor prognosis.The etiology and pathogenesis of the disease are still unclear.The current reports in the literature are related to the imbalance of multiple cytokines and signal pathways.The name of the disease in TCM is "Yin Yang Du".In recent years,more and more studies have been done on the theory and pathogenesis of this yin-yang-toxicity.Lupus erythematosus is a series of diseases,with cutaneous lupus erythematosus with only skin lesions and systemic lupus erythematosus with multiple organs and multiple systems involvement.This study only focused on skin lesions of lupus erythematosus.Skin and systemic skin lesions were included.ObjectiveTo investigate the expression of JAK2/STAT3 in lesions of skin and observe its expression level.At the same time comparing the expression differences between normal skin tissue to hope to further explore their role in the pathogenesis of lupus erythematosus and provide new ideas and theoretical basis for clinical treatment.Previous studies of lupus erythematosus have been less concerned with cytokines and signaling pathways such as JAK2/STAT3.This study uses "yin and yang" in the theory as a syndrome differentiation program and the signal pathway JAK2/STAT3 as a research indicator.The differences in the expression levels of JAK2,STAT3 and its phosphorylated P-JAK2,P-STAT3 between yin toxin syndromes and yang toxin syndromes can be hoped that the modern significance of the theory of yin and yang toxicity can be further discussed.Discussing the role of this signaling pathway in the pathogenesis and development of lupus erythematosus to provide clues for further elucidating the pathogenesis of lupus erythematosus developing,molecular diagnostics and drug target.The Chinese medicine can be used as a basis for the micro-differentiation of"Yin and Yang",further enriching the traditional Chinese medicine theory research and inspiring new experimental ideas.MethodsIn the experiment,22 cases of lupus erythematosus skin lesions and 10 normal control skin tissues were detected by immunohistochemistry.In the experimental group,symptoms were divided into two groups:the serotoxic group,the 10 cases and the sinus poisonous group,and 12 cases.The differences in the expression of JAK2,STAT3 and their phosphorylated P-JAK2,P-STAT3 were detected.The images were analyzed using image analysis system software and the positive results were evaluated using the average optical density values.The expression of each protein was compared between the experimental group and the normal skin group and the difference between the Yin group and the Yang group.Using SPSS20.0 statistical software for data processing.All data is represented by (?)±s.The significance between the groups was compared with the normal distribution of data using analysis of variance and t-test.Wilcoxon Rank Sum Test for data not conforming to normal distribution.Both tests will use two-sided inspection.The test level was a=0.05,and P<0.05 was considered statistically significant.The result1 JAK2,P-JAK2,STAT3,and P-STAT3 all showed moderate to strong positive expression in the cytoplasm and nucleus of epidermal cells of the patient’s lesion,and were extremely weakly positively expressed in normal skin epidermis.There were significant differences(P<0.01).2 JAK2,STAT3,P-JAK2,and P-STAT3 showed moderate to strong positive expression in the serotonergic group and weakly positive expression in the sinus toxin group.There was a significant difference between the two groups(P<0.01).3 JAK2,STAT3,P-JAK2,and P-STAT3 expressed positively in the experimental group and showed very weak positive expression in normal skin.There was a significant difference between the two groups(P<0.01).4 JAK2,STAT3,P-JAK2,and P-STAT3 were weakly to mid-positive lesions in the sinister group and showed very weak positive expression in the normal skin tissue.There was a significant difference between the two groups(P<0.01).5 Correlation analysis,because the upstream of STATs is JAKs,that is,JAK2/STAT3 signal pathway has a certain correlation,this experiment also confirmed this point.5.1 The expression of JAK2 and STAT3 in LE skin lesions was linearly correlated(pearson correlation coefficient r=0.761,P=0.000<0.01).5.2 P-JAK2 and P-STAT3 expression in LE lesions was linearly correlated(pearson correlation coefficient r=0.752,P=0.000<0.01).5.3 The expression of JAK2 and P-JAK2 was linear in LE skin lesions(pearson correlation coefficient r=0.659,P=0.001<0.01).5.4 The expression of STAT3 and P-STAT3 in LE lesions was linearly correlated(pearson correlation coefficient r=0.780,P=0.000<0.01).Conclusion1.JAK2/STAT3 and its phosphorylated P-JAK2,P-STAT3 showed moderate to strong positive expression in the cytoplasm and nucleus of epidermis full-thickness cells in lesions of lupus erythematosus,but no expression in normal skin tissues.There was a significant difference between the two,suggesting that JAK2/STAT3 is involved in the inflammatory process of lupus erythematosus.2.The expression of JAK2,STAT3 and its phosphorylated P-JAK2,P-STAT3 were positively correlated in the lesions of patients,suggesting that there may be direct or indirect regulation between the three.3.There was a significant difference between the positive group and the negative group of P-JAK2 and P-STAT3 in the serotoxic group.This can be used to explore the microscopic differentiation of yin and yang toxicity theoretically,and the phosphorylation of P-JAK2 and P-STAT3 represents Cell damage can further explore the theories of yin and yang and explain where "poison" comes from. |
