| Atherosclerosis(AS)is a systemic,chronic,progressive disease that is arterial lipid deposition,endometrial hyperplasia of fibre calcification in the artery wall thickening and hardening,the process of luminal stenosis,coronary heart disease,cerebrovascular disease and thromboembolic disorders such AS the main pathological basis of ischemic cardio-cerebrovascular disease.In a wide range of traditional risk factors(such as hypertension,diabetes,hyperlipidemia,smoking,etc.),the study more and more attention to the Homocysteine(Hcy)and the correlation of atherosclerosis.High homocysteine levels(Hyperhomocysteinemia,HHcy)is an independent risk factor for atherosclerosis,high homocysteine levels on the growth of endothelial cells and endothelial dysfunction plays a role of resistance or damaged endothelial change again,affect the normal endothelial diastolic function,also can quicken the speed of neointimal hyperplasia of thickening,smooth muscle cell proliferation and synchronization is accelerated,thus cause endothelial dysfunction.Moxibustion therapy has been used for thousands of years.It ignites moxa or other herbs and directly or indirectly stimulates body surface points or specific parts through burning,smoking,etc.,with the help of the mild heat of moxibustion and the drug effect of smoke.Through the conduction of meridians and acupoints,warm ventilation,active removal of sputum,solid prospering,and positive spondylosis,a traditional external treatment method for disease prevention,treatment,rehabilitation and health care is achieved.Previous clinical trials of this group have demonstrated that moxibustion can reduce serum Hcy levels in hypertensive patients,and after a series of experiments on atherosclerosis in animals,it was concluded that moxibustion may affect blood lipids,coagulation and fibrinolysis systems,and inflammation of blood vessels.Reactions,the function of vascular endothelial cells and many other ways protect the cardiovascular system.Based on the current research foundation and research progress at home and abroad,the research group will use moxibustion and moxa smoke as an intervention method to feed C57BL/6J mice with a high methionine diet as a modeling method and observation object.Moxibustion and moxa smoke in the treatment of HHcy and its preventive mechanism for early AS.OBJECTIVEObserve the effect of moxibustion and moxa smoke on hyperhomocysteinemia,and from the perspective of enhancing antioxidant capacity and protecting vascular endothelium,explore the role of early intervention of moxibustion and moxa smoke in the prevention and treatment of atherosclerosis.METHODSA total of 53 8-week-old SPF healthy female C57BL/6J mice were divided into four groups:blank group,model group,moxa smoke group,and moxibustion group;8 blank groups,normal diet,and the rest of each group 15 only,feed with high methionine feed.Moxibustion group:The mice were grasped and placed into a fixture to fix the head,tail and limbs of the mouse.Put the mouse into the holder and set it up.There is a small hole at the bottom of the holder.The hole in the chest is exposed and the worm is placed under the chest of the mouse.The heat is transmitted through the hole in the bottom of the holder to the chest of the mouse.In the process of moxibustion,the height of moxa bars is constantly adjusted so that the temperature of the moxibustion remains basically constant.Moxibustion intervention 20min/time(20min/time,1 time/day,6 times/week),Moxa smoke group:High methionine feed,the mice were captured,fixed,the same method with the moxibustion group,and then the holder was placed in a homemade glass jar,lit moxa,using a dust meter to detect the concentration of smoke in the glass jar,so that moxa smoke concentration was kept at 10-15 mg/m3(20 min/time,1 time/day,6 times/week).Model group:high methionine feeding,grabbing and fixing mice,method with moxibustion group,no other intervention;Blank group:normal diet feeding,no other intervention.The mice were sacrificed after 12 weeks of intervention.RESULT1.Serum levels of Hcy and SAH:Model group,moxibustion group,and Moxa smoke group were all significantly higher than the Blank group;compared with the model group,the levels of Hcy and SAH in the moxibustion group were significantly reduced,and there was no significant difference in the moxa smoke group.However,there is a downward trend.2.Contents of aorta SOD,eNOS,ox-LDL and HO-1:Compared with the model group,SOD and HO-1 in the aorta of the moxibustion group were significantly increased,ox-LDL was significantly decreased,and eNOS had no significant difference;The group HO-1 was significantly higher than the model group,and the other indicators were not statistically different.3.Aorta and liver pathological observation:compared with the blank group,the thoracic aorta of the model group showed severely disordered vascular endothelium,a large number of foam cells accumulated,and the fiber arrangement was lacking in continuity;endothelial dysfunction was found in the moxibustion group and the smoke group.Foam cell extracellular lipid accumulation,poor fiber continuity.Hepatic pathology showed swelling and disorganized cells.There were obvious lipid droplets in the model group.There were scattered lipid droplets in the moxibustion group and Moxa smoke group.The arrangement of cells was more regular.CONCLUSIONHigh methionine diet can cause hyperhomocysteinemia.After a period of intervention,it can cause oxidative stress and injury of arterial endothelium.Through moxibustion intervention,serum Hey and intermediate metabolites SAH content can be reduced,SOD and HO-1.can be increased,ox-LDL content can be reduced,and it can enhance the ability of the body to resist oxidation and protect endothelial function.As one effective factors of moxibustion,moxa smoke can also reduce Hey content to a certain extent,reduce HO-1,and show some antioxidant capacity.Compared with model group,SAH,ox-LDL and SOD express certain the trend,but the results were no significant differences,the overall control effect is not as obvious as the moxibustion group. |
