Ligusticum chuanxiong Hort.was a traditional Chinese medicine which comes from Ligusticum,Umbrella family.In this paper,we use ultrasonic assisted extraction of volatile oil and polysaccharides to study how different drying methods impacted the physicochemical properties and antioxidant activities of polysaccharides on Ligusticum chuanxiong.Also we try to figure out the effects of the antioxidant and antibacterial activities of volatile oil of Ligusticum chuanxiong.The main research results are as follows:1.Using ultrasonic assisted extraction assay to research the assisted extraction process of volatile oil and polysaccharide from Ligusticum chuanxiong.The results showed that the extraction rate of volatile oil and polysaccharide of Ligusticum chuanxiong could be promoted by the ratio of liquid to material,extraction time andultrasonic time to a certain extent.The optimal extraction process of volatile oil and polysaccharide from Ligusticum chuanxiong was as follows:the ratio of liquid to material was 15.85mL/g,the extraction time was 4.27h and the ultrasonic time was 16.45min.Under this condition,the predictive value of extraction rate of volatile oil was 1.32%,polysaccharide extraction rate was 2.64%.Taking into account the convenience of the test operation,we verified the optimal conditions,and the result were as follows:the ratio of material to liquid was 16 mL/g,the extraction time was 4.3h and the ultrasonic time was 17min.Then,performing the experiment for 3 times,the corresponding extraction rate of volatile oil was 1.35%,the extraction rate of polysaccharide was 2.72%,and the relative error was 2.2%and 3.0%,respectively..It is shown that the optimal condition parameters obtained by the optimization of the response surface design are reliable and can predict the test yield better.2.The effects of different drying methods(hot air drying,vacuum drying and freeze drying)on the physicochemical properties and antioxidant activity of crude polysaccharides of Ligusticum chuanxiong were studied.As the results shown,the content of total polysaccharide and protein in LCP-F was the highest,and the content of uronic acid in LCP-V was the highest in vacuum drying.In the research of physical and chemical properties,we found that there were no significant differences in the physical and chemical properties of different drying methods.Moreover in the research of antioxidant activity,the effect of different drying methods on its antioxidant activity was remarkablelly significant.The antioxidant activity of LCP-F was higher than that of LCP-V and LCP-H.3.The antioxidant activity of volatile oil of Ligusticum chuanxiong showed that DPPH free radicals,· OH radicals and 02-· free radicals all had the ability to scavenge all kinds of free radicals,which means it had good antioxidant activity.Furthermore,the ability to cavenge all kinds of free radicals all increased as the concentration of the sample increased within the experimental concentration range.The order of strong free radical scavenging ability of volatile oil of chuanxiong was DPPH radical>· OH radical>02-· free radicals.4.The results of antibacterial activity in vitro showed that the Ligusticum chuanxiong volatile oil on Staphylococcus aureus,Bacillus subtilis,Pseudomonas aeruginosa and Escherichia coli all had larger antibactierial circle than TBHQ.That indicated that the volatile oil of Ligusticum chuanxiong has a better antibacterial effect on Staphylococcus aureus,Pseudomonas aeruginosa,Escherichia coli.Volatile oil on Staphylococcus aureus and E.coli were less than 0.5%,while the MIC of Bacillus subtilis and Pseudomonas aeruginosa were less than 0.8%.According to that,the inhibitory effect of volatile oil of Ligusticum chuanxiong on Staphylococcus aureus and Escherichia coli was better than that of Bacillus subtilis and Pseudomonas aeruginosa.Besides,volatile oil of Ligusticum chuanxiong had no apparent inhibitory effect on salmonella.
