Phosphorus?P?is one of the most important nutrient elements in the growth and development of organisms such as plants,microorganisms,etc.,and is the key component of cell membranes,it participate in the physiological and biochemical process of biology in various ways,plays an important role in promoting biological metabolism,growth and development.Phosphate transporter proteins were used to mediate the absorption and transshipment of phosphorus in organisms such as plants and mycorrhizal fungi,etc..Based on the latest whole-genome sequencing results of 8kinds of higher Macrofungi which were Boletus edulis,Laccaria bicolor,Agaricus bisporus,Trametes versicolor,Pholiota nameko,Tuber melanosporum,Schizophyllum commune and Rhizopogon luteolus.in the paper,by using computer technology and bioinformatic analysis method,amino acid sequences and corresponding nucleic acid sequences of 19 phosphate transporter PHO84 were compared and analyzed in order to understand deeply the properties,structure of these PHO84.These above will provide theoretical basis for establishing the reasonable structure of phosphorus nutrition on artificial domestication of mycorrhizal fungi and high yield cultivation of mushroom etc.,for further understanding the mechanism of phosphorus transport and use of the mycorrhizal fungi and their host plants.The main research conclusions are as follows:1.To 8 kinds of macrofungi,19 PHO84 sequences,which had 5 in L.bicolor,4 in A.bisporus and T.versicolor respectively,2 in S.commune,1 in B.edulis,P.nameko,T.melanosporum and R.luteolus respectively,were totally compared in NCBI database using the protein series of phosphate transporters PHO84?NP013583.1?of Saccharomyces cerevisiae S288C6 as a query.2.The average length of 19 PHO84 genes was around 1786 bp,their GC?Lb PHO84-2,Lb PHO84-3,Lb PHO84-4,Lb PHO84-5?,?Ab PHO84-2,Ab PHO84-3,Ab PHO84-4?and 1 in Pholiota nameko?Pn PHO84?,was lower than AT content,which were similar to the PHO84 genes of Saccharomyces cerevisiae.3.Analyzed the physical and chemical properties of the amino acid residues of,the number,the molecular weight,the number of atoms,etc..The physicochemical properties of 19 PHO84 proteins such as amino acid residue type,quantity,molecular weight,atomic number,etc..were analyzed,and the results showed that the proportion of non-polar aliphatic amino acid?Alanine?in T.versicolor Tv PHO84-4 was the highest?12.14%?,and polarity neutral amino acid?cysteine?in T.versicolor Tv PHO84-1 was the lowest?0.54%?.the number of amino acid residues of T.versicolor Tv PHO84-2 was the most,their molecular weight and atomic number were 68723.23 u and 9700 respectively;19 proteins were all hydrophobic proteins,whose half-life was 30 h,and in which the basic protein was most,whose theoretical isoelectric point was 7.96-9.20.4.The phosphorylation sites of 19 PHO84 proteins were mainly focused on serine residues,followed by threonine and tyrosine residues,and were mainly distributed at both ends of primary structure of protein polypeptide chain.5.19 PHO84 proteins had not been found signal peptide;and subcellular localization analysis showed that they were all located in the cell membrane.6.The analysis of the secondary structure of 19 PHO84 protein showed that?-helix accounted for about 40%,followed by random coil?26%?and extended strand?24%?,the rest was beta-rotation?About 10%?.7.19 PHO84 proteins contained 912 transmembrane domains,and their conservative structures were basically similar.8.The synonymous codon,their use parameters and parameters correlation analysis showed that the average GC contents of 19 PHO84 gene was 53.7%,which indicated that synonymous codon was biased towards the use of two nucleotides of guanine and cytosine.the effective number of codons?ENC?was higher?the mean value was 50.8?,and their corresponding endogenous gene expression was lower;the codon adaptation index?CAI?were 0.126 to 0.326,indicating that the gene have weak bias to the codon;GC content had significant and extremely significant correlation with ENC,CAI etc.,GC3s?the contents of G+C in the third synonymous codon?had negative significant correlation with ENC,CAI etc.,which indicated that Guanine significantly affected the gene expression level,then affected the preference of the synonymous condon.9.Two phylogenetic trees constructed with RSCU and amino acid sequence had more difference,but the first clustering at a similar distance could be completed among intraspecies of T.versicolor,intraspecies and interspecies of A.bisporus and L.bicolor,which was consistent with the traditional classification at certain degree;because genes and proteins are the relationship of upstream and downstream,more perfect clustering methods need to be further verified by combing the study of transcriptome and proteome etc.in the future.
