Optical coherence tomography is a non-invasive,non-radiative,high-resolution,high speed and promising optical imaging technology with extensive application in the fields of biology and medicine.Besides the structure information,polarization sensitive optical coherence tomography(PSOCT)can provide additional polarization information of the sample,which is promising for early diagnosis of diseases.This dissertation introduces the development,principle,system performances,and main applications of OCT The theory and principle of PSOCT technology is demonstrated,and the basic PSOCT systems are introduced.Based on the spectral domain OCT system developed in the lab,a set of fully single-mode fiber,single-spectral PSOCT system was proposed and developed.A polarization-sensitive linear wavenumber spectrometer based on Wollaston prism was designed and built.A polarization-based adjustment method based on polarization controller was implemented.The retardation and optical axis of the standard compensator and waveplate were accurately measured.The depth-resolved intensity images,retardation images and optical axis images of burdock tissue and chicken muscle tissue were used to verify the feasibility of the polarization-sensitive OCT system for acquiring biological tissue polarization information.A fundus OCT imaging system based on Graphic Processing Unit(GPU)image reconstruction was designed and built.Utilizing the parallel computing function of GPU,the CUDA C language is used to realize the rapid processing of OCT data.The OCT sample arm is designed based on the frame of a slit lamp.The depth-resolved intensity image,retardation image and optic axis image was obtained.A fiber-optic endoscopic polarization-sensitive swept source optical coherence tomography system was designed.A fiber optic rotary connector for the endoscopic OCT system was developed.The rotary connector adopts a brushless DC servo motor to drive rotation,and the high-efficiency coupling of light energy is realized by injecting a refractive index matching liquid in a glass capillary.
