Phellinus pini is a white-rot fungus and Basidiomycetous fungus belonging to Hymenochaetaceac.It can cause decay of trees,so it is great harmful to trees.A large number of research shows that Phellinus has good bioactivity activites,such as anti-tumor properties,antioxidant and antiviral.While the study of Phellinus pini was scarce,so the thesis was aimed to provides clues for the further development and utilization of Phellinus pini.(1)This paper first studied the optimization of technological conditions of microwave extraction of polysaccharides and polyphenol from Phellinus pini.Through single factor and response surface experiment design method,the result shows that the optimal extraction conditions of polysaccharides is 1:40 ratio of water to material,extraction temperature 92?,extraction time 60min.under the optimized conditions,the experimental value of polysaccharides yield is 3.33%.The result shows that the optimal extraction conditions of polyphenol is 1:20 ratio of water to material,ethanol concentration 55%,extraction temperature 42?,extraction time 42min.under the optimized conditions,the experimental value of polyphenol yield is 3.98%.(2)Secondly,the antioxidant activities and inhibitory activities of a-glucosidase are assessed with the polysaccharides and polyphenol extracts under the optimized conditions.The result shows that polysaccharides and polyphenol in the certain concentration range,both have certain antioxidant activities and a-glucosidase inhibitory activities.The DPPH EC50 Value of polysaccharides and phenolics was 2.59 and 1.03 mg/mL,separately;The ABTS EC50 Value of polysaccharides and phenolics was 6.44 and 2.26 mg/mL,separately;and the total antioxidant capacity of polysaccharides and phenolics was 0.27 and 0.13 mmol FeS04/mg,separately.The a-glucosidase inhibitory activity(IC50)of polysaccharides and phenolics was 0.15 and 2.17mg/mL,separately.Both a little lower than the value of 4.26mg/mL,a positive control of acarbose.(3)Finally,under the optimized conditions,the polysaccharides fraction from water extraction of Phellinus pini after ethanol precipitation with the concentration of 60%is isolated and purified using DEAE Sepharose Fast Flow and Sephacryl S gel-filtration chromatography repeatedly.PHP60A and PHP60B1 are obtained.Through HPLC detection,the result indicats that both are homogeneous and their molecular weight are 88.9 KDa and 178.6 KDa.PHP60A and PHP60B1 are investigated by sugar analysis,PHP60A and PHP60B1 are composed of mannose,galactose and glucose with the molar ratio of 0.86:4.89:1.00 and 0.74:4.53:1.00 respectively.
