In this paper,a small dose of poorly soluble drugs vinpocetine,was used as a model drug.The technology of solid dispersion was used to improve the low solubility and bioavailability of vinpocetine and enhance its speed of dissolution.Then osmotic pump tablets of vinpocetine solid dispersion was prepared to improve patients'compliance,reduce fluctuation of the plasma concentration and improve drug safety.Meanwhile,the single factor study was researched about the osmotic pump tablets of vinpocetine solid dispersion,and the design-response surface methodology was applied to optimize the formulation prescription for the foundation of animal experiments.The analysis method of UV spectrophotometry was studied to measure the content and release degree in vitro of vinpocetine solid dispersion and its osmotic pump tablets.Methodological study results were consistent with the requirements.In this paper,PEG6000,PVP K30 and F68 were as the material.By adjusting the ratio of vinpocetine and the material,vinpocetine solid dispersion were prepared,which could significantly improve the dissolution rate and dissolve rate.Then the thermodynamic was investigated.Results indicated that,citric acid?a cidifying agent?,could improve dissolution and stability of vinpocetine solid dispersion some extent;Dispersion process of vinpocetine in PVP K30 was an exothermic process.The driving force was the enthalpy-driven.Rising temperature was adverse to the formation of solid dispersions.In this paper,homemade vinpocetine solid dispersion was as a main drug.By adjusting the formulation rate of the tablet core and the film-coated,osmotic pump tablets of vinpocetine solid dispersion with a certain release rate were prepared.The single factor study of preparation prescription results showed that,the first factor affected the drug release was the amount of osmotic promoting agent in a tablet core;Meanwhile,the content of PEG in a coating liquid and the weightening of film coating were also factors.Based on results of the single factor experiments above,the formulation prescription of osmotic pump tablets of vinpocetine solid dispersions was optimized by central composite design-response surface methodology,with the amount of penetration enhancers in a tablet core?X1?,the content of PEG in coating solution?X2?and the weight gain after drying?X3?on the responses of the correlation coefficients of 0-12h release profiles?Y1?,and cumulative release amount within 12 hours?Y2?.Then optimized prescription was verificated and consisted with results.The final results showed that the predicted value was basically consistent with actual values,and proved that mathematical model had a good forecasting effect.
