Research On Isolation,Screening Of The Active Straims And Exploring Secondary Metabolite Constituents Of Chinese Medicine Plants Endophytic Actinomycete Of Sichuan

Thanks to the unique natural geographical environment,sichuan province has "library" of traditional Chinese medicine(TCM),Chinese herbal medicine amount and type in the country.Long-term since,the wisdom of the human by the animal and plant natural products to treat disease.But some Chinese herbal medicine itself defects lead to unable to industrial production.And some Chinese herbal medicine itself defects lead to unable to industrial production.Therefore,by modern science and technology to seek new methods for natural products is of great significance.This study to gulin county,sichuan authentic Chinese herbal medicine from yellow grass as the research material,through the separation of pure culture technology access to 118 strains actinomycetes.In all strains by 16 s rRNA gene sequencing and homology sequence alignment,fully shows that the cast yellow grass endophytes rich diversity,they respectively belong to five Suborders:Suborder Glycomycineae,Suborder Micromonosporineae,Suborder Streptomycineae,Suborder Streptosporangineae,Suborder Corynebacterineae.And belong to 12 generas:Streptomyces,Streptomonospora,Catellatospora,Microbispora,Micromonospora,Actinomadura,Glycomyces,Virgisporangium,Nocardiopsis,Streptosporangium,Nocardia,Rhodococcus.Using by golden brown staphylococcus,shame scale mycobacteria,fluorescent pseudomonas,saccharomyces cerevisiae,red yeast,e.coli and other pathogenic bacteria as indicator bacteria strains of appeal preliminary selection,breeding good strains with an activity(Z11(ZHP4055))as a large number of experimental fungus fermentation.On the basis of the optimal fermentation conditions were determined for subsequent fermentation culture.By resin absorption of 108 g,10 kinds the compounds were isolated by,respectively by the wave of the franco-prussian identification,they are Phenylacetic acid,Cyclo(L-Pro-D-Met),2,5-Piperazinedione,Cyclo(L-Pro-L-Val),DL-proline.
