Gel electrophoresis is mainly used for the detection and analysis of biological macromolecular.There is indispensable and complicated dyeing and decolonization process in the analytic process of slab gel electrophoresis,but it is time-consuming.However the capillary gel electrophoresis can shorten the analysis time effectively.In terms of biological macromolecules detection,gel electrophoresis is still a common method of detection,but the literature related to the analysis of small molecules is rarely reported.Chip electrophoresis is a fast and efficient electrophoresis analysis technology.The detection and analysis of amino acid is one of the important research of analytical chemistry.There is complicated derivatization process in amino acids with optical detector.So this thesis will combine conductive coupling detector with column gel electrophoresis or chip electrophoresis,finally,label-free amino acids can be detected and analyzed online.In the first chapter,the principle and detection mode of gel electrophoresis used to analyze to biological molecules is described.The controlling of electroosmotic flow or joule heat in capillary electrophoresis and chip electrophoresis preparation materials are introduced in detail.What's more,the development of conductivity detection which is used to analyze amino acid is summarized.In the second chapter,with the help of the column gel system,mixed amino acids have been separated and detected successfully.Injection of sample,voltage of separation,effective detection distance and the choice of sample matrix have been optimized in the electrolyte solution of acetic acid.Under the condition of optimal experiment,the standard curve of amino acid has also been drawn.In the third chapter,Label-free amino acids are separated and detected online in the chip electrophoresis.In the 3.2 mollL acetic acid-0.05%tween-20 supporting electrolyte solution,the detection of separation and the reproducibility analysis of six mixed amino acids are implemented,and the detection limit reaches the grade of ?mol/L.Chip electrophoresis coupled with conductivity detection device presents potential applications in the detecting of non-labeled biological molecular.Electrophoresis detection device in this work is simple.It is easy to operate and low-cost,and is promising to detect bio-macromolecule.
