Obesity has become one of the most important factors harming human health in the world in the 21 st century.It is the main predisposing factor of diseases such as hyperglycemia,hypertension,hyperlipidemia,insulin resistance and atherosclerosis.Obesity not only affects people’s quality of life,but also increases the financial burden.Elaeagnus mollis oil is a kind of vegetable oil extracted from the seeds of Elaeagnus mollis Diels.It is unique and precious,and rich in nutritional components,which has physiological effects,such as anti-inflammatory,antioxidant,blood lipid lowering and so on.Aerobic exercise is considered to be a scientific and effective way to lose weight.It is welcomed by people because of moderate intensity,limited space,diverse forms and participation by all.The purpose of this study was to investigate the intervention of Elaeagnus mollis oil combined with aerobic exercise in decreasing obesity and reducing lipid levels in mice with high-fat diet.Four-week-old male C57BL/6 mice were used.After one week of acclimation on laboratory chow,they were randomly divided into the 5groups(n=8): normal control group(NC),high-fat diet group(HFD),Elaeagnus mollis oil group(EMO),aerobic exercise group(AE),Elaeagnus mollis oil combined with aerobic exercise group(EMO/AE).NC group was fed with normal diet,other groups were fed with high-fat diet,EMO group was gavage with Elaeagnus mollis oil(3.03g/kg·d).AE group taken moderate aerobic exercise.The exercise prescription was 6 times per week,progressing to 60 min/session.EMO/AE group was gavage with Elaeagnus mollis oil and taken moderate aerobic exercise.During 8 weeks of intervention,the food intake and growth status were recorded every day,and the body weight of mice was measured weekly.The mice were killed by cervical dislocation under anesthesia,and the body length was measured,serum,liver and adipose tissue were collected.Serum TC,TG,HDL-C and LDL-C in bloodwere measured using commercial kits and AI was calculated.The pathological changes of liver and adipose tissue were observed by pathological section.The expressions of SREBP-1c,FAS,ACC and ACOX m RNA in liver tissue and HSL,LPL and UCP-1 m RNA in adipose tissue were detected by real-time fluorescence quantitative PCR.The results showed that:(1)The body weight,Lee’s index,fat weight,fat coefficient,serum TC,TG,LDL-C and AI of EMO group,AE group and EMO/AE group were significantly lower than those of HFD group(P<0.05).The fat weight,fat coefficient,serum TC,TG,LDL-C and AI of EMO/AE group were lower than those of EMO group and AE group.Results showed that Elaeagnus mollis oil,aerobic exercise and the combined group inhibited the body weight gain of mice with high fat diet,and reduced Lee’s index,fat weight and coefficient,and improved the level of blood lipid.The combined intervention showed better effect.(2)Compared with the HFD group,the number of adipocytes and hepatocytes of the EMO group,AE group and EMO/AE group decreased,and the hepatic lobule structure of the EMO group,AE group and EMO/AE group partially recovered,and the lipid droplet vacuoles in liver tissue significantly decreased.The effect of EMO/AE group was more obvious.Results showed that Elaeagnus mollis oil,aerobic exercise and the combined group reduced the size of fat cells in adipose tissue and the lipid droplet vacuoles in liver,leading to inhibit fat accumulation,and improve liver lesions.The combined intervention showed better effect.(3)Compared with the HFD group,the expression of SREBP-1c,FAS and ACC m RNA of the EMO group,AE group and EMO/AE group decreased,and the expression of ACOX m RNA increased in liver.In adipose tissue,the expression of HSL and LPL m RNA of EMO group,AE group and EMO/AE group was significantly higher than that of HFD group(P<0.05)partly.The expression of UCP-1 m RNA of AE group and EMO/AE group was significantly higher than that of HFD group(P<0.05).Results showed that Elaeagnus mollis oil,aerobic exercise and the combined group down-regulated the expression of SREBP-1c,FAS and ACC m RNA in liver,and up-regulated the expression of ACOX m RNA in liver and HSL,LPL and UCP-1 m RNA in adipose tissue,leading to improve fat deposition and fat metabolism in liver,and reduce fat accumulation in vivo.In summary,Elaeagnus mollis oil,aerobic exercise and the combination of Elaeagnus mollis oil and aerobic exercise had the effect of decreasing obesity and lipid-lowering levels in mice with high-fat diet.The combined intervention showed better effect.
