Blumea DC.is one of the most important genera of the Inuleae taxonomic group of the Asteracea family.Most of the genera are annual or perennial aromatic plants.As a traditional Chinese medicine with a long history,this genus is used as a tea and seasoning.It has biological activities such as deodorization,anti-inflammatory,insecticide,anti-tumor,anti-obesity,sterilization,and antioxidation.However,the chemical composition of this genus is complex,and the effective ingredients and mechanism of action have not been fully clarified.The functional components of this genus need to be further explored to promote the research and development of this genus.This thesis systematically develops the extraction and composition analysis of essential oils from the genus Blumea DC.in China,as well as the research on the antioxidation,anti-tumor,anti-inflammatory and antibacterial biological activities of essential oils.The main findings are as follows:Using steam distillation(SD)method,simultaneous distillation extraction(SDE)method,and headspace solid-phase microextraction(HS-SPME)method,six species of Blumea DC.(Blumea oblongifolia,Blumea aromatica,Blumea megacephala,Blumea lanciniata,Blumea riparia,and Blumea hieracifolia)essential oils were extracted,the chemical composition of the essential oils was identified by gas chromatography-mass spectrometry,and the yields of essential oils extracted from different parts of the plants by SD method and SDE method were evaluated.The results showed that the extraction of essential oils by SD,SDE and HS-SPME has a large difference in composition,which may be due to the temperature and separation patterns of different extraction methods;the use of SDE method to extract six plants essential oils had a higher yield than SD method;However,the SD method and SDE method were more consistent in the yields of essential oils extracted from different parts of the plants.The content of essential oils in the Blumea oblongifolia,Blumea aromatica,Blumea megacephala,and Blumea lanciniata was higher in the leaves.The content of essential oils in the Blumea riparia and Blumea hieracifolia was higher in the stem.Cluster analysis of the main components of 6kinds of plants essential oils could be divided Blumea aromatica,Blumea riparia,Blumea oblongifolia,and Blumea hieracifolia intoⅠ,Blumea megacephala and Blumea lanciniata were classified intoⅡ.The biological activities study on the essential oils extracted by the SDE method showed that the essential oils of this genus had a low certain antioxidation ability,and had limited scavenging ability to DPPH,OH,O2-free radicals,and the FRAP value of the experimental concentration of essential oils samples were low.The anti-tumor experiment found that when the concentration of essential oils was less than 50 mg/L,the inhibition rate of LO2cells was less than 10%;the inhibition of proliferation of Hep G2 cells did not exceed 13%,and the inhibition rate of proliferation of H1299 cells increased in a concentration-dependent manner.It tended to be stable,and the overall inhibitory effect showed that the essential oils group was lower than 5-fluorouracil in the positive control.The anti-inflammatory activity showed that within the range of 3.125-50μg/m L,6 kinds of essential oils had no toxic and side effects on the growth of RAW264.7 cells.And the Blumea lanciniata essential oils and Blumea hieracifolia essential oils had significant inhibitory effect on the release of NO,IL-1,IL-6 and TNF-ɑinduced by LPS in RAW264.7 cells(P<0.05),and could also inhibit the transcription of IL-6,i NOS and TNF-ɑm RNA in cells to different degrees,thus effectively inhibiting the occurrence of inflammation..The antibacterial activity experiment found that the essential oils of Blumea megacephala was highly sensitive to Escherichia coli,and the Blumea lanciniata essential oils was highly sensitive to Staphylococcus aureus,and these two essential oils could effectively cooperate with antibiotic to inhibit drug-resistant S.aureus strains(120882,130149,132166);the research on its antibacterial mechanism showed that the synergistic effect of essential oils and antibiotic could lead to the leakage of drug-resistant bacteria protein and the change of cell membrane permeability,thus inhibiting the normal growth of drug-resistant bacteria;Blumea lanciniata essential oils could cause intracellular electrolyte and protein leakage,the integrity of the cell membrane was destroyed,thereby inhibiting the normal growth of S.aureus.
