In this paper,the fish roe proteins were used as the main raw materials,including atlantic salmon,crucian carp,anchovy,Japanese eel,and carp.In silico enzymatic hydrolysis of the protein was performed by online database.And then,the bioactivity,solubility,and toxicity of the released peptides were predicted using online program.Molecular docking was used to screen di-and tri-peptides which strongly bonded with acetylcholinesterase(AChE),Bubutyrocholinesterase(BChE)andβ-amyloid precursor proteolytic enzyme 1(BACE1).Besides,the interaction mechanism of the peptide with the enzyme were evaluated by Discovery Studio.At last,the whole factory design was completed.The present study provides a theoretical basis for the utilization of fish roe protein resources and the application of bioactive peptides.The main research results are as follows:1.The online tool ExPASy PeptideCutter was used to release the peptide from the roe protein of atlantic salmon,crucian carp,anchovy,Japanese eel,and carp.the PeptideRanker,Innovagen,and ToxinPred programs were used to predict the bioactivity,solubility and toxicity of the 102 di-and tri-peptides.Combined with molecular docking,four tri-peptides WIR,MIR,FQK and YPR with in vitro anti-Alzheimer’s(AD)activity were identified.Among the four peptides,peptide WIR shows the activity against AChE(IC50 value of 43.32±1.22μM),BChE(33.5%inhibition rate at 1.06±0.87 mM),and BACE1(IC50 value of 2.27±0.35 mM).2.The tri-peptides WIR,MIR,FQK and YPR mainly formed hydrogen bond interactions with the amino residues,i.e.,Tyr334,Tyr70,Tyr121 and Trp279 of AChE,hydrogen bond interactions with the amino residues,i.e.,His438,Trp82,Asp70,and Ser198 of BChE.And these four peptides mainly formed hydrogen bonding interactions with the amino residues,i.e.,Trp115,Ile110,Asn233,Thr232,Asp228,Asp32,Tyr71,and Phe108 of BACE1.3.The 10 tons of fish roe peptide pilot plant design was completed.The production plan of the peptide was designed,and the equipment was selected.It was estimated that the electricity consumption was 4454.41 kWh/shift,and the water consumption was 78.64 t/shift.The economic benefit analysis of 10 tons fish roe peptides was carried out.The annual net profit was RMB 1.6366 million and the annual profit rate was 16.4%,per year.
