During the fermentation of Luzhou-flavor liquor Daqu,Daqu largely determines the brewing quality.Different Daqu contains different enzyme-producing microbial community structure and biodiversity.Using molecular biology methods.The experimental conclusions obtained are as follows:(1)The colony morphology of prokaryotic producing bacteria producing pectinase was 9 types,and the colony morphology of eukaryotic producing bacteria was 10 types.Molecular biology identification of prokaryotic microorganisms producing pectinase belongs to 6 genera and belongs to the genus Bacillus.The eukaryotic microorganisms producing pectinase include Aspergillus(3 species)and Penicillium(1species)and 4 species,of which Aspergillus is the dominant bacterial group.In addition,the prokaryotic microorganism that is not reported to produce pectinase is Bacillus paralicheniformis strain.(2)There are 12 types of colony forms of prokaryotic microorganisms producing xylanase,and 10 types of colony forms of eukaryotic microorganisms.It has been identified that there are 10 genera of two genera(Bacillus and Cronobacter)of prokaryotic microorganisms producing xylanase,among which there are 8species of Bacillus genus,showing rich diversity.There are four genera of xylanase-producing eukaryotic microorganisms,namely Aspergillus(2 species),Lichtheimia(2 species),Rhizomucor(1 species),and Penicillium(1 species).Among them,Bacillus oryzaecorticis strain,Bacillus paralicheniformis strain and Lichtheimia ramosa strain are rarely reported.(3)Among cellulase producing bacteria,there are 13 types of prokaryotic microbial colonies and 8 types of eukaryotic microbial colonies.Bacillus(5 species)and Cronobacter(1 species)were identified as prokaryotic microbial strains producing cellulase.Four genera were identified from cellulase-producing eukaryotic microbial strains,namely Lichtheimia(2 species),Mycocladus(1 species),Penicillium(1 species)and Rhizopus(1species).The eukaryotic microorganism Lichtheimia ramosa strain producing xylanase has not been reported.(4)Among the amylase-producing bacteria,there are 11 types of prokaryotic microbial colonies;and 8 types of eukaryotic microbial colonies.According to the results of species identification,there are a total of 6 genera of Bacillus(5 species)and Aneurinibacillus(1 species)in the prokaryotic microorganism strains producing amylase.Bacillus is the predominant bacterial group among prokaryotic microorganisms producing amylase.There are four genera of amylase-producing eukaryotic microorganisms,namely Aspergillus(3 species),Rhizopus(1 species),Mycocladus(1 species)and Mucor(1 species).Among the amylase-producing microorganisms identified in the experiment,Bacillus glycinifermentans strain and Mycocladus corymbiferus strain were reported for the first time in this study.(5)There are 10 types of prokaryotic microbial colonies producingsaccharification enzymes,and 7 types of eukaryotic microbial colonies.Screening identified only one genus of prokaryotic microorganisms producing saccharification enzymes,Bacillus(4 species).A total of three genera and three species were identified in eukaryotic microorganisms producing saccharification enzymes,namely Lichtheimia,Mycocladus and Aspergillus.Molecular biology has identified the saccharification enzyme-producing eukaryotic microorganism Mycocladus corymbiferus strain,which is rarely reported.The research on the microbial enzyme system in Daqu through modern biotechnology will lay a solid foundation for the research and development of enhanced Daqu and Daqu enzyme preparations,the improvement of liquor brewing process,the improvement of liquor quality,and the vigorous development of China's liquor industry.
