| Objective: To observe the effect of different electroacupunctures on intestinal flora activity and BDNF expression in hippocampus of depression model rats,and to explore the mechanism of electro acupuncture on depressive symptoms.Methods: The rats were randomly divided into normal group,the CUMS model group,including music group,the pulse electric acupuncture group and the fluoxetine group,duplicate the CUMS model,after the success of the building to the different intervention methods,the comparison of each rat behavior assessment.The intestinal contents of rats were extracted for biolog-eco anaerobic culture,and the average color change rate(AWCD)and Shannon index of intestinal flora of rats were detected by Biolog system.Brain tissue sections in the hippocampal area were conducted,and BDNF positive cells in the hippocampal tissue of rats were detected by immunohistochemistry.Results(1)Compared with the normal group,the hair of the model group lost its luster,the diet decreased,and the weight difference was obvious(P < 0.05).Compared with rats in the model group,the weight of fluoxetine group,music electroacupuncture group and pulse electroacupuncture group all increased significantly(P < 0.05).There was no significant weight difference between fluoxetine group and music electroacupuncture group(P > 0.05).The weight gain of music electroacupuncture group was compared with that of pulse electroacupuncture group(P < 0.05).(2)AWCD values of all groups reached the highest value after incubation for 72 h and tended to be stable.AWCD values of the model group were significantly lower than those of the normal group(P < 0.05).Compared with CUMS model group,the drug group,music electroacupuncture group and pulse electroacupuncture group were significantly increased(P < 0.05).Among them,music electroacupuncture group was slightly higher than pulse electroacupuncture group,with no significant difference(P >0.05),and significantly higher than fluoxetine group,with statistical significance(P < 0.05).(3)The comparison between groups of rat hippocampus BDNF expression changes,compared with normal group,the total number of BDNF positive cells CUMS model group decreased significantly(P < 0.05),arrange the sparse and cell vacuolated,mostly cytoplasm staining obviously lighter than normal group.The number of cells in fluoxetine group,music electroacupuncture group and pulse electroacupuncture group was significantly higher thanthat in the model group(P < 0.05),and the cytoplasmic staining was significantly better than that in the model group.Among them,the total number of BDNF positive cells in the music electroacupuncture group was slightly higher than that in the pulse electroacupuncture group,showing no statistical significance(P > 0.05),and significantly higher than that in the fluoxetine group,showing a significant difference(P < 0.05).Conclusions(1)Music electroacupuncture,pulse electroacupuncture and fluoxetine intervention can all improve behavioral performance of depressed rats.Among them,music electroacupuncture has the same effect as fluoxetine and is superior to pulse electroacupuncture.(2)Different electricity can regulate the activity of intestinal flora and the expression of BDNF in hippocampus of depression model rats,and it is better than fluoxetine.(3)One of the mechanisms by which different electroacupuncture may interfere with depressive symptoms may be to achieve therapeutic purposes by regulating intestinal flora activity and hippocampal BDNF. |
