| Objective:To observe the impact of gypsum rice decoction on yeast-induced rats tumor necrosis factor-α(TNF-α),Cytokine interleukin1β(IL-1β),Cytokine interleukin 6(IL-6),Ca2+,NF-κBp65 and COX-2.And try to discuss the mechanisms of gypsum rice decoction and provide experimental basis for clinical use.Methods:Randomly selected 10 from 50 Male SD rats weight(180±20)g to normal control group,the rest of 40 were injected with 20%yeast suspension under 10ml/kg to establish models of fever.After 4h of the model,the rats was successfully established fever model.Then we randomly divided the model rats into four groups:10 in model group,10 in aspirin group,10 in gypsum rice decoction group,10 in gypsum decoction group.Aspirin group,gypsum rice decoction group and gypsum decoction were given corresponding decoction intragastric administration according to the dose of0.1g/kg,7.83g/kg,17.62g/kg respectively.The normal group and model group were given isometrical normal saline intragastric administration.After injecting yeast,the anal temperature was measured 1time per 1h,and 10 times were measured.The temperature of rats at different time points was recorded and the maximum temperature rise height(ΔTmax)and the temperature response index of 6h(TRI6)were calculated.Enzyme-linked immunosorbent assay was used to detect the level of serum TNF-α,IL-1β,IL-6,Ca2+in each group.Hematoxylin-eosin staining was used to observe the histopathological changes in hypothalamus of rats.Immunohistochemistry and RT-qPCR were used to detect the expression of NF-κBp65,COX-2 in hypothalamus of each group.SPSS.22 statistics software was used to analyze the data.Results:1.Temperature changes in rats:After the administration of 13h,the temperature increased gradually in the rest of the groups except the normal group.Compared with the model group,Aspirin group’s temperature difference decreased,the difference had statistical significance(P<0.01),there were no significant differences in gypsum rice decoction group and gypsum decoction group(P>0.05,P>0.05).After administration of 46h,aspirin,gypsum rice decoction and gypsum decoction could inhibit the increase of temperature of febrile rats,showing the antipyretic effect.And aspirin’s antipyretic effect was better than gypsum rice decoction and gypsum decoction,the difference had statistical significance(P<0.01,P<0.01).there were no significant differences between the gypsum rice decoction and gypsum decoction(P>0.05).2.The expression level of TNF-α,IL-1β,IL-6,Ca2+in serum:After administration of 6h,the level of serum TNF-α,IL-1β,IL-6 in aspirin group,gypsum rice group and gypsum group were all over lower than model group,the difference was statistically significant(P<0.01,P<0.01,P<0.01).Compared with model group,gypsum rice decoction could increase the content of Ca2+in serum,the difference were statistically significant(P<0.01,P<0.01).3.HE staining in hypothalamus:Compared with normal control group,the neurons in model group suffered from pyknosis,vacuoles,edema,swelling and inflammatory cell infiltration.After administration of 6h,aspirin group,gypsum rice decoction and gypsum decoction had alleviated pathological changes in hypothalamus.4.the expression level of NF-κBp65 protein in hypothalamus:the expression level of NF-κBp65 protein in hypothalamus in aspirin group,gypsum rice decoction group and gypsum decoction group were all lower thanmodelgroup,thedifferencewerestatistically significant(P<0.01,P<0.01,P<0.01).5.the expression level of COX-2 mRNA in hypothalamus:the expression level of COX-2 mRNA in hypothalamus in aspirin group,gypsum rice decoction group and gypsum decoction group were all lower than model group,the difference was statistically significant(P<0.01,P<0.01,P<0.01).Conclusion:Gypsum rice decoction has antipyretic effect in yeast-induced rats,the mechanism of antipyretic effect may be related to the decrease of TNF-α,IL-1β,IL-6 in serum and the increase of Ca2+in serum,and inhibit the expression of NF-κBp65 protein and COX-2 mRNA in hypothalamus. |
