Objective:1.Establish BN rats' DR laser surgery models.2.To investigate the effects of Danhuang mingmu decoction on DR laser postoperative models of BN rats and the expression of VEGF/PEDF.Methods:1.Part 1:Establish BN rats' DR laser surgery models.Using 40mg·kg-1 STZ intraperitoneal injection to induce DR model of BN rats.After STZ injection,blood glucose was stable to 16.7mmol/L and above,thus,the DM models were established.After 3 months,Measure blood glucose,weight and observe the lens condition,and FFA examination and p-selectin,cAMP,cGMP were performed and the retinal tissue morphology was observed after HE dyeing under light microscopy,confirming the successful establishment of DR model.All DR model rats had laser photocoagulation in right eye and left eye without laser photocoagulation.,and the FFA was examined after one week.2.Part 2:Investigate the intervention of Danhuang Mingmu decoction on DR laser postoperative models of BN rats.All the BN rats' right eyes of the model group carry out laser surgery,and the left eyes had not been carried out laser photocoagulation.Then,the model group was randomly divided into three groups:B,C and D,and the blank group was group A,each group having 10 BN rats.Group A and group B were filled with distilled water to the stomach,group C was hydroxyl benzene sulfonic acidcalcium suspension lavage 5 ml/kg(concentration of 100 ml,containing hydroxyl benzene sulfonic acid calcium 1000 mg),group D was Danhuang Mingmu decoction 5 ml/kg to fill the stomach,are equivalent to the adult equivalent dose.After 4 weeks,the blood glucose and P-Selectin,cAMP,cGMP of the BN rats were measured,and the retinal tissue was observed under the staining light microscope,and the expression of VEGF and PEDF was detected by Western blot.Results:Part 1:1.After 3 months molding,the lens of the blank group was not obviously cloudy,and the structure of the fundus was clear.However,part of the lens of the model group was cloudy,and the bottom of the eye was not clear.2.After 3 months modeling,the blood glucose of the model group was significantly higher than that in the blank group,P<0.05,and the difference was statistically significant.3.After 3 months modeling,FFA was employed to check the visual base of the model group with microhemangioma,and the blank group did not see the changes in the fundus of microangioma.4.After 3 months modeling,the content of P-selectin in the plasma of the model group was higher than that in the blank group,P<0.05,and the difference was statistically significant.5.After 3 months modeling,cAMP and cGMP of the model group were higher than the blank group,P<0.05,and the difference was statistically significant.6.After 3 months modeling,the retina structure of the blank group was more disordered than that of the model group.7.After laser photocoagulation of DR model rats,the laser class was seen in the model group.Part 2:1.4 weeks after gavage,group A weight significantly heavier than the other three groups,and the difference was statistically significant(P<0.05).The weight of group B,group C,group D has no difference.(P>0.05).2.After 4 weeks of gavage,the blood glucose of group A was significantly lower than that of the other three groups,P<0.05,and the difference was statistically significant.The comparison among group B,group C and group D,P>0.05,showed no statistical difference.3.After 4 weeks of gavage,the content of P-selectin in group A was the lowest,and P-Selectin in group B was the highest.Group A was compared with group B and group C,P<0.05,and the difference was statistically significant.There was no statistical difference between group A and group D.(P>0.05).4.After 4 weeks of gavage,the concentration of cAMP and cGMP in group B and group C were higher than that in group A and group D,P<0.05,and the difference was statistically significant.Compared group A with group D,P>0.05,and the difference has no statistically significant.5.After 4 weeks of gavage,the relative expression of VEGF was detected by WB,and the relative gray value of group B and group C were the highest,and has statistically significant compared with group A and C(P<0.05).Compared group A with group D,P>0.05,the difference was no statistically significant.6.After 4 weeks of gavage,the relative expression quantity of PEDF was detected by WB,and the relative gray value of group B and group C was relatively low,and compared with group A and C,P<0.05,the difference was statistically significant.Compared group A with group D,P>0.05,the difference was no statistically significant.Conclusion:1.The DR model and DR after laser surgery model of BN rats were characterized by blood stasis,Yin deficiency and Yang hyperactivity.2.Danhuang Mingmu decoction can improve the blood stasis index of DR rats.3.Danhuang Mingmu decoction can improve the index of Yin deficiency and Yang hyperactivity in DR rats.4.Danhuang Mingmu decoction can be used to repair the retinal of theBN rat DR after laser photocoagulation.5.Danhuang Mingmu decoction can reduce the relative expression of the retinal VEGF and improve the relative expression of retinal PEDF of DR after laser photocoagulation in BN rats.Meanwhile,VEGF and PEDF were antagonistic factors.6.It is speculated that the possible mechanism of Yangyin Qingre of Danhuang Mingmu decoction in the treatment for DR after laser surgery may by reducing the relative expression of VEGF,and rasing the relative expression of PEDF...
