Gastric injury is a stomach disease caused by multiple factors together.Excessive drinking is one of the most important factors in the pathogenesis.Hericium erinaceus is a traditional medicinal and edible fungi,which is mainly used for the treatment of digestive tract diseases including gastric ulcer,acute and chronic gastritis.Based on the protective effect of H.erinaceus on the gastrointestinal tract,this study tried to evaluate the effect of H.erinaceus powder on mice with acute and chronic ethanol-induced gastric injury and explore its mechanism.The main research is as follows:(1)To explore the preventive effect of H.erinaceus powder on ethanol-induced acute gastric injury in mice.In this study,mice were given acute alcoholic gavage as a model,and mice were given different doses of H.erinaceus powder(125 ~ 500 mg/kg)for 6 days.Then a single gavage of anhydrous ethanol was used.We investigated the preventive effect of H.erinaceus powder on ethanol-induced acute gastric injury by measuring the bleeding,edema,cell necrosis and inflammatory cell infiltration in gastric mucosa.We also measured the pH of gastric juice,the change of malondialdehyde(MDA)in stomach tissue,the TNF-α,IL-1β and MPO mRNA expression and the content of inflammatory proteins Caspase-1,NLRP3,IL-1β,TNF-α and TLR4.The results showed that H.erinaceus powder could significantly improve the hemorrhage,ulceration,cell necrosis and shedding of gastric mucosa in acute gastric injury mice.Compared with the model group,the scores of histomorphological changes of gastric mucosa in the low,medium and high dose groups were reduced by 16.93%,19.52% and 42.89%,respectively.Compared with the model group,the expression levels of TNF-α mRNA in gastric tissues of the low,medium,and high dose groups decreased by 49.02%,54.64% and 63.10%,respectively;the expression of IL-1β mRNA decreased by 27.79%.,43.87%,48.26%;MPO mRNA expression in the high dose group decreased by 42.69%.This indicated that H.erinaceus powder could significantly reduce the mRNA expression of TNF-α,IL-1β and MPO in gastric tissue.It can also reduce the production of inflammatory proteins Caspase-1,NLRP3,IL-1β,TNF-α and TLR4 and reduce the degree of inflammation.However,the impact on gastric juice pH and MDA is limited.(2)To investigate the therapeutic effect of H.erinaceus powder on ethanol-induced chronic gastric injury in mice.The experiment was based on chronic gastric injury mice with alcohol feeding plus a single alcohol consumption.Mice were feed with low-dose ethanol continuously for 10 days.Then a single gavage of 63% ethanol was given.Different doses of H.erinaceus powder(0.5 ~ 1 g/kg)were administrated.We measured the bleeding,edema,cell necrosis,inflammatory cell infiltration,and the mRNA expression of TNF-α,IL-1β,and MPO in gastric mucosa,and the content of inflammatory proteins Caspase-1,NLRP3,IL-1β,TNF-α and TLR4 to investigate the effect of H.erinaceus powder on the ethanol-induced chronic gastric injury.The results showed that H.erinaceus could significantly improve bleeding,edema,inflammatory cell infiltration,epithelial cell shedding in gastric mucosa of chronic gastric injury mice.Compared with the model group,the tissue morphology scores of the low and high dose group of H.erinaceus powder were decreased by 63.61% and 74.55%.The expression levels of TNF-α mRNA in the low and high dose group were decreased by 45.84% and 43.67%;that of IL-1β mRNA were decreased by 37.59% and 34.83%;and that of MPO mRNA were decreased by 33.43% and 42.16%.This indicated that H.erinaceus powder could significantly reduce the mRNA expression of TNF-α,IL-1β,and MPO in gastric tissue,to achieve the effect of treating gastric injury.It can also reduce the production of inflammatory proteins Caspase-1,NLRP3,IL-1β,TNF-α and TLR4 and reduce the degree of inflammation.(3)To explore the mechanism of H.erinaceus powder in treating ethanol-induced chronic gastric injury in mice.Proteins in CTL,ETOH,and HEH were extracted from the stomach tissue of the mice and quantified by iTRAQ.Compared with the database,a total of 5842 proteins were obtained.We obtained 308,205,and 230 differential proteins in ETOH vs.CTL,HEH vs.CTL,HEH vs.ETOH,respectively.The proteins were annotated and GO enrichment was conducted.We found that the protein involved in keratinization,epidermal cell differentiation,keratinocyte differentiation,epidermis development,cornified envelope,and intermediate filament were enriched.The KEGG signaling enrichment was conducted.It was found that the protein involved in the p53 pathway was enriched,suggesting that the p53 pathway may affect the gastric mucosal status.It is speculated that H.erinaceus powder may treat chronic gastric injury by affecting the differentiation of gastric epithelial cells.
