| ObjectiveIn this study,apoptosis index and the expression levels of the apoptosis related factors like Bax,Bcl-2,Fas,Fas-L and P53 were detected in three differenct FGR rat models.We aim to explore the apoptosis characteristics of placental cells induced by different factors,and provide novle evidence for the classification and intervention of FGR.Methods1.Animal Models:There are three rat models such as low-protein diet intake of FGR rat model,the nicotine injection of FGR rat model and the idiopathic of FGR rat model had been established in previous experiment.2.The apoptosis index in the placental tissues of these four groups were observed and counted by TUNEL.3.The expression level of apoptotic factors(Bax,Bcl-2 and P53)in the mitochondrial parthway of apoptosis in placental cells was both detected by Real-time quantitative PCR and immunohistochemical.4.The expression level of apoptotic factors(Fas and Fas-L)in the death receptor parthway of apoptosis in placental cells was both detected by Real-time quantitative PCR and immunohistochemical.Results1.Compared to the control group,the low-protein group and the nicotine group have significant growth restriction with fetal rat,markedly decreased average birth weight and increased the incidence of FGR(*P<0.05).2.After the apoptosis index of the placental tissues of these four groups were observed and counted by TUNEL.It is concluded that the apoptosis index in the Basal Zone is significantly higher than in the Labyrinth Zone(P<0.01).Compared to the control group,the apoptosis index of the low-protein group,the nicotine group and the idiopathic group were significantly increased(P<0.01).3.The expression level of apoptotic factors in the mitochondrial parthway of apoptosis in placental cells was both detected by Real-time quantitative PCR and immunohistochemical.3.1 Compared to the control group,the expression of Bax,Bcl-2 and P53 have no significant difference in the low-protein group.3.2 Compared to the control group,the expression of Bax,Bcl-2 and P53 have significant difference in the nicotine group.3.3 Compared to the control group,the expression of Bax,Bcl-2 and P53 have significant difference in the idiopathic group.4.The expression level of apoptotic factors in the death receptor parthway of apoptosis in placental cells was both detected by Real-time quantitative PCR and immunohistochemical.4.1 Compared to the control group,the expression of Fas and Fas-L have significant difference in the low-protein group.4.2 Compared to the control group,the expression of Fas-L have significant difference in the nicotine group.4.3 Compared to the control group,the expression of Fas-L have significant difference in the idiopathic group.ConclusionThere are significant differences between these three types of FGR rat models and normal pregnant rats,and caused apoptosis up-regulated of placenta,limit growth of fetal rats by different pathways. |
