Objective:Ginger is a rhizome of Gingeraceae,an herbaceous perennial monocotyledonous ginger.As a widely used traditional Chinese medicine herbs,has been version of"Chinese Pharmacopoeia"included,spleen and stomach Indications,dizziness and vomiting,vomiting more than blood,is commonly used traditional Chinese medicine.In this paper,ginger,ginger and ginger plant volatile oil components,trace elements,chemical composition,the determination of which the main component of 6-gingerol content for the research and quality control of ginger ingredients to provide the basis for the study and MOE and other software The main component of 6-gingerol efficacy prediction,and related efficacy experiments,to explain the basis of its efficacy material,and pharmacological research provide the basis for the study.Methods:Volatile oils from each species of Zingiber officinale were extracted by steam distillation,the chemical constituents were analyzed by gas chromatography-mass spectrometry?GC-MS?after diluting the essential oil with diethyl ether,and the results were compared.Eight species of Zingiberaceae wet nitrification,and then use atomic fluorescence spectrophotometer to determine the content.Using ethanol reflux extraction method,taking the content of 6-gingerol and the rate of ointment as indexes,the orthogonal experiment was carried out on the four factors of extracting times,extracting time,alcohol concentration and adding amount of alcohol.ZINGIBERIS RHIZOMA 10.0 kg,70%ethanol extracted in the concentration tank 3times,each 2 hours,the combined concentrate.The concentrate was extracted with petroleum ether,dichloromethane,ethyl acetate,n-butanol and concentrated to give an extract.The fractions of petroleum ether,methylene chloride and ethyl acetate were separated and purified by repeated silica gel column chromatography and ODS column chromatography.According to the physical and chemical properties of the isolated monomer compounds and PIP(1H-NMR,13C-NMR)data were analyzed to identify the structure of the compound.Establishment of mouse ear swelling model,to explore the anti-inflammatory effect of ginger extract.In vitro diffusion experiments were conducted with the TP-6 horizontal diffusion cell.The isolated rat skin was selected as the osmotic barrier and sampled from the receiving cell.The content of 6-gingerol in the sample was determined by HPLC.The use of MOE software to explore 6-gingerol target prediction and molecular docking,the foundation for the future mechanism of the study.Results:Eight kinds of ginger plant volatile oil out of the rate of 0.0075-0.06 ml/g,identified chemical constituents were dried ginger 75,turmeric 52,Curcuma 71,galangal 80,76 83 of nutmeg,45 of nutmeg and 67 of Amomum villosum.There were significant differences in the contents of heavy metal elements in the samples,including Hg,galangal,zedoary turmeric,dried ginger,Ginger>dried ginger,galangal,etc.>strawberry,and its content is lower than the national standard of heavy metal content;Hg:recovery rate:85.77%-111.52%,RSD%:0.35%-1.07%,As:89.92-119.92%,RSD%:0.42%-1.13%,which indicates that the method has the advantages of high accuracy,simple operation,good stability and reproducibility,and can be used for the determination and analysis of heavy metal elements in different medicinal materials.Through data processing and the actual situation,The optimal extraction technology of 6-gingerol in dried ginger was determined as follows:adding 16 times the amount of 90%ethanol,extracting for 2 hours and extracting for 3 times,the average content of 6-gingerol was 127.13 mg/g;Six compounds were identified and identified as follows:8-gingerol?1?,10-gingerol?2?,stigmasterol?3?,?-sitosterol,6-gingerol?5?and carotasterol?6?.The ear edema weight and ear swelling in the group of 5 g/kg of ginger extract were significantly lower?P<0.05?Anti-inflammatory effect;6-gingerol can penetrate the isolated rat skin;30%ethanol,3%azone on 6-gingerol significant role in promoting penetration.Conclusion:There are great differences in the components of the volatile oil between different Zingiberaceae plants,and the zingiberaceae plants are also different in their genus,but each plant chemical composition has its own uniqueness and its special chemical composition.Orthogonal design can the extraction conditions of 6-gingerol in Ginger were optimized conveniently and accurately.The optimized extraction conditions were reproducible,the extraction rate was high and the process was reasonable.The separation and identification of the chemical constituents of Ginger were the best choice for Ginger,Ginger extract has a certain anti-inflammatory effect,which lays the foundation for the further study of its components;This study will provide experimental evidence for the preparation of 6-gingerol transdermal delivery preparation;Through the target Prediction and molecular docking,to explore the possible efficacy of 6-gingerol,and lay the foundation for further mechanism studies.
