Objective:The aim of this study was to investigat the long term effects of high voltage electric burn on serum interleukin-6,interleukin-3,β-platelet globulin and the intervention effect of Xuebijing,ulinastatin.Our aims was to study the changes of serum IL-6,IL-3,β-TG in rats with high voltage electrical burns,and to study the molecular mechanism of cytokine injury and microcirculation disturbance after burn injury through intervention of Xuebijing,ulinastain.We want to investigate the feasibility of Xuebijing and ulinastain in the treatment of high voltage electric burn.Methods:We divided randomly three hundred and fifty SD rats into fake high-voltage electrical burns group(control group),high-voltage electrical burns group,high-voltage electrical burns saline treatment group,high-voltage electrical burns Xuebijing treatment group,high-voltage electrical burns UTI treatment group,each group contained 70 rats.According to the observation time,we divided rats in each group randomly.That was 15 minutes before injury,1hour after electric injury,8h,24h,48h,72h,1 week after electric injury,10 rats in each group.According to the experimental requirements,we established the rat model.Rats in the control group received the same device but no electricity.The corresponding drug intervention was given at regular intervals according to the group and the time phase.Saline control group by2ml/kg intraperitoneal injection of saline,Xuebijing group by 10ml/kg intraperitoneal injection of Xuebijing,Ulinastatin group by 2×10~4u/kg(2ml/kg)intraperitoneal injection of ulinastatin And we administrated abdominally the corresponding drugs regularly,except for the fake high-voltage electrical burns group(control group)and the electric injury group.According to the time of experimentation,we collected collection and preservation of the specimens and store the serum in a refrigerator at-70℃.Apply ELISA double antibody method to detect the levels of IL-6,IL-3,andβ-TG in serum.Results:1 IL-6 contents in electrical injury groups are generally higher than those in unelectrical groups(P<0.05),and IL-6 contents within the 6 phases injury showed significant differences(P<0.05),showing a rising trend,the level of content in 1h,8h,24h,48h,72h,1week after injury is higher than the value of this group before injury(P<0.05).IL-6 contents in xuebijing groups salt groups are generally lower than those in salt groups(P<0.05),and IL-6content over time after the injury showed significant differences(P<0.05),the level of content in 1h,8h,24h,48h,72h,1week after injury showed significant differences(P<0.05),The IL-6 contents rises to the highest value(60.62±10.24)pg/ml at 8 hours after electrical injury,Down to the lowest level(44.63±10.17)pg/ml 1 week after electrical injury.IL-6 contents in UTI groups are generally lower than those in salt groups(P<0.05),and IL-6content over time after the injury showed significant differences(P<0.05),the level of content in 1h,8h,24h,48h,72h,1week after injury showed significant differences(P<0.05).The IL-6 contents rises to the highest value(73.40±7.47)pg/ml at 8 hours after electrical injury,Down to the lowest level(53.84±9.52)pg/ml 1 week after electrical injury.There was no significant difference(P>0.05)between the two groups in 24 hours,48hours,72 hours and one weed after injury,indicating that saline also had a therapeutic effect on high voltage burns.2 The expression of IL-3 in the electrical injury groups was higher than that in the unelectrical group,and the IL-3 contents in the electrical injury group was affected by the time variable(P<0.05),the level of content in 1h,8h,24h,48h,72h,1week after injury is higher than the value of this group before injury(P<0.05).The IL-3 content in the this group was lower than that in the saline group(P<0.05),and the xuebijing group affected by the time variable(P<0.05),The IL-3 contents rises to the highest value(65.76±9.45)pg/ml at1 hours after electrical injury,Down to the lowest level(40.90±5.62)pg/ml one week after electrical injury.There was no significant difference(P>0.05)between the two groups in 48 hours,72 hours and one week after injury,indicating that saline also had a therapeutic effect on high voltage burns.The IL-3 content in the UTI group was lower than that in the saline group(P<0.05),and the UTI group affected by the time variable(P<0.05).The IL-3 contents rises to the highest value(65.15±9.34)pg/ml at 8 hours after electrical injury,Down to the lowest level(41.00±7.76)pg/ml one week after electrical injury.There was no significant difference(P>0.05)between the two groups in 24hours,72 hours and one week after injury,indicating that saline also had a therapeutic effect on high voltage burns.3 The expression ofβ-TG in the electrical injury groups was higher than that in the unelectrical group,and theβ-TG content in the electrical injury group was affected by the time variable(P<0.05),the level of the content in 1h,8h,24h,48h,72h,1week after injury is higher than the value of the group before injury(P<0.05).Theβ-TG content in the xuebijing group was lower than that in the saline group(P<0.05),and the xuebijing group affected by the time variable(P<0.05),There is a significant difference of the content level between the unelectrical group and the electrical greep.Theβ-TG contents rises to the highest value(16.26±1.42)pg/ml at 1 hours after electrical injury,Down to the lowest level(10.19±2.09)pg/ml one week after electrical injury.There was no significant difference(P>0.05)between the two groups in 72hours and one week after injury,indicating that saline also had a therapeutic effect on high voltage burns.Theβ-TG content in the UTI group was lower than that in the saline group(P<0.05),and theβ-TG group affected by the time variable(P<0.05).Theβ-TG contents rises to the highest value(21.26±1.81)pg/ml at 8 hours after electrical injury,Down to the lowest level(10.71±0.88)pg/ml one week after electrical injury.There was no significant difference(P>0.05)between the two groups in 48 hours,72 hours and one week after injury,indicating that saline also had a therapeutic effect on high voltage burns.Conclusions:The levels of IL-6,IL-3andβ-TG in serum of rats were significantly increased after high voltage burn.Xuebijing injection and UTI could inhibit the secretion of IL-6,IL-3 andβ-TG,reduce the damage of cytokines,improve the state of microcirculation,improve body immunity and promote wound healing.
