| Objective:To investigate the differences in expression and clinical significance of DDR1 and ASPP2 in tongue squamous cell carcinoma(TSCC)and its adjacent non tumor tissues.Methods:45 specimens of tongue cancer and 30 non paracancerous tissue specimens(from cancer tissue > 1.5cm)were collected from Department of Stomatology,Yantai Yuhuangding Hospital,Qingdao University(from January 2014 to March 2017),and take them as object of study.Immunohistochemical method was used to detect the expression of DDR1 and ASPP2 protein.Chi-square test was used to analyze the relationship between the two and clinical pathological characteristics,and the correlation between DDR1 and ASPP2.Results:(1)The positive rate of DDR1 and ASPP2 in tongue squamous cell carcinoma was significantly higher than that of non tumor tissue.The positive rate of the tongue cancer tissue and adjacent non tumor tissues were 53.33% and 13.33%(χ2=12.310,P=0.001)in turn.At the same time,the positive rate of ASPP2 were 57.78% and 6.67%(χ2=20.099,P=0.000).The difference of rate is obvious and has statistical significance.(2)The expression of DDR1 protein was related to the degree of differentiation,lymph node metastasis,but do not have statistical significance with sex,age,history of smoking and clinical T stage.The positive rate of DDR1 protein was about 40.63% in well-differentiated tissue of pathological differentiation,and about 84.62%(P < 0.05)in the moderately and poorly differentiated tissues.The positive rate in the lymph node metastasis group was about 85.71%,and that in the non lymph node metastasis group was about 40%(P < 0.05).(3)The expression of ASPP2 protein is only related to the degree of pathological differentiation,and is not related to other clinical and pathological parameters.In degree of pathological differentiation,the positive expression rate of ASPP2 protein in well-differentiated group was about 46.88%,and that was 84.62% in moderately and poorly differentiated group,the difference was statistically significant(P < 0.05).(4)There were 24 cases of DDR1 positiveiy expressed in 45 cases of tongue cancer,of which 18 cases were positive for ASPP2 co expression.In 30 nontumor tissues adjacent to cancer,13 cases were negative about the expression of DDR1 and ASPP2 protein.After Spearman rank correlation analysis,the two was positive correlation(P =0.012,gamma =0.373).Conclusion:(1)The expression of DDR1 protein is related to the differentiation degree and lymph node metastasis,but not related to sex,age,history of smoking and clinical T stage.Therefore,DDR1 is closely related to tongue cancer,and plays an important role in the development and metastasis of tongue cancer.(2)The expression of ASPP2 protein is only related to the degree of pathological differentiation,and is not related to other clinical and pathological parameters.This shows that ASPP2 is involved in the process of the development of tongue cancer and has an important impact on it.(3)In tongue squamous cell carcinoma,there is a positive correlation between the expression of DDR1 and ASPP2 protein,and there may be some mutual promotion between the two in occurrence and development of tongue cancer.So,detecting the two expression simultaneously,is expected to become a molecular marker for clinical diagnosis,malignancy judgement and prognosis of tongue cancer,and it may provide new direction for targeted therapy of cancer. |
