Objective: To observe the effect of Shengshanzha Decoction on blood lipids and hemorheology related factors in rats with impaired glucose tolerance,and to explore the therapeutic effect and mechanism of raw Hawthorn fruit and herbs on prediabetes,so as to provide experimental basis for further research on traditional Chinese medicine diet health theory,And provide new ideas and methods for clinical prevention and treatment of diabetes.Methods: A total of 70 SPF-grade SD rats were randomly selected as the normal control group and fed with common feed.The remaining rats were fed a high-fat diet.After 3 weeks of feeding,fasting for 12 hours,dissolving STZ in a 1% buffer with citric acid buffer,and intraperitoneal injection of STZ 30 mg/kg once by weight of the rat was completed within 30 minutes.After 3 days fasting overnight,blood was taken from the tail vein to measure fasting blood glucose(FPG).Then,each group was treated with 1 ml/100 g of 20% glucose solution,and 2 hours later,blood was taken from the tail vein to measure blood glucose.With FBG at 6.1mmol/L ~ 7.0mmol/L,2h postprandial blood glucose ? 7.8 mmol/L,but <11.1 mmol/L was determined to be successful in the rat model of impaired glucose tolerance.Fifty model rats with abnormal glucose tolerance were randomly divided into five groups: abnormal glucose tolerance model group,high-dose Radix Scutellariae group,middle-dose Radix Sanguisorba,low-dose Radix Mossanii,and metformin group,10 in each group.Another 10 normal rats in the same batch were used as the normal group.The normal group and the model group were intragastrically administrated with the same volume of normal saline.The rest of the groups were given drugs according to the “equivalent dose ratio table of humans and animals converted according to body surface area”,and the high,middle,and low doses of Hawthorn were weighed at 1000 g per day.4g,2g,and 1g of raw Hawthorn were separately administered to the stomach;in the metformin group,0.125 g of suspension was administered by gavage at a dose of 1000 g once a day for 3 weeks.During the dosing period,the normal group was fed with common feed.The model group,high dose group of raw Hawthorn,middle dose group of Health Hawthorn,low dose group of Hawthorn and metformin were fed with high-fat diet,and the body weight,activity,and feeding condition of the animals were monitored.,deaths,etc.,weekly fasting blood glucose measured 1 times.After 3 weeks,7 animals were randomly selected from each group and fasted for 12 hours.Blood was taken from the abdominal artery and blood glucose,blood lipids,and hemorheology were measured.Differences among groups were compared by one-way ANOVA.Results: Compared with the normal group,the model group cholesterol,triglyceride,low density lipoprotein cholesterol,whole blood high cut relative index,whole blood low cut relative index,whole blood high cut reduction viscosity,whole blood low cut reduction viscosity,fasting Blood glucose increased(P<0.01)and high-density lipoprotein cholesterol decreased(P<0.01).Compared with the model group,the Metformin group,Shengshanyiyin high-,middle-,and low-dose group cholesterol,triglyceride,low-density lipoprotein cholesterol,whole blood high-cutting relative index,whole blood low-cutting relative index,whole blood high cut reduction Viscosity,low-cut reduction viscosity of whole blood,and fasting blood glucose all decreased in varying degrees,while high-density lipoprotein cholesterol in metformin group and high-,medium-,and low-dose group of Shengshan Qiyin increased to varying degrees.For triglycerides,low-density lipoprotein cholesterol,and high-density lipoprotein cholesterol,the highest dose of Shengshan Quyin was most obvious(P<0.01).For cholesterol,whole blood high cut relative index,whole blood low cut relative index,whole blood high cut reduction viscosity,whole blood low cut reduction viscosity,and fasting blood glucose,the differences between the metformin group and the Shengshanyi drink large,medium,and small dose groups No statistical significance(P>0.05).Conclusion: Shengshanyiyin can prevent and treat the development of impaired glucose tolerance by lowering triglycerides,cholesterol,low-density lipoprotein and increasing high-density lipoprotein content,intervening in disorders of lipid metabolism,and reducing blood viscosity to reduce blood glucose.This not only confirms the fact that raw Hawthorn,which has the same origin of traditional Chinese medicines and medicines,but also has the advantages of promoting blood circulation and removing blood stasis,has a definite curative effect in preventing and treating impaired glucose tolerance,and provides an experimental basis for the traditional Chinese medicine diet health theory.
